扶肾降浊方对系膜增生性肾炎肾小管间质损害大鼠COLⅠ及COLⅢ蛋白和基因表达的影响

摘　　要：目的观察扶肾降浊方对系膜增生性肾炎大鼠肾小管间质细胞外基质Ⅰ、Ⅲ型胶原(COLⅠ及COLⅢ)蛋白和基因的调节作用。方法采用系膜增生性肾炎模型,将大鼠随机分为正常组、模型组、扶肾降浊方中药组、贝那普利西药组。采用免疫组化法和实时荧光定量PCR法分别检测实验12周、16周和20周末各组Ⅰ、Ⅲ型胶原蛋白和基因的表达。结果 12周、16周和20周末的模型组肾脏COLⅠ及COLⅢ蛋白表达呈强阳性(P<0.01),COLⅠ及COLⅢ基因表达上调(P<0.01),扶肾降浊方中药组和贝那普利组大鼠肾脏COLⅠ及COLⅢ蛋白表达均较模型组降低(P<0.01),COLⅠ及COLⅢ基因表达亦均较模型组下调(P<0.05);随着造模时间的延长,模型组肾小管COLⅠ及COLⅢ蛋白和基因表达均呈升高趋势,治疗组仍作用明显(P<0.05)。结论扶肾降浊方可通过基因和蛋白水平调节COLⅠ和COLⅢ在肾脏的表达,减少COLⅠ和COLⅢ在肾小管间质的积聚以减轻肾间质纤维化,从而对肾小管间质损害起到防治作用。Objective To observe the regulatory effect of＂Fushen Jiangzhuo Formula＂on the protein and gene expression of Collagen type Ⅰand Collagen type Ⅲin tubulointerstitial extracellular matrix in rats with mesangial proliferative glomerulonephritis. Methods Using mesangial proliferative glomerulonephritis model,rats were randomly divided into normal group,model group,Chinese medicine group,and Benazepril group. We detected the protein expression and gene expression of COLⅠ and COLⅢ of each group for in 12th week,16th week and 20th week chinese medicine group with immunohistochemistry experiments and realtime fluorescence quantitative PCR. Results After 12 weeks,16 weeks and 20 weeks,the protein expression of COLⅠ and COLⅢ on kidney in the model group were strongly positive（ P〈0. 01）,the gene expression of COLⅠ and COLⅢ on kidney increased（ P〈0. 01）,the protein expression of COLⅠ and COLⅢ on kidney in the TCM group and benazepril group reduced significantly（ P〈0. 01） compared with the model group,and the gene expression of COLⅠ and COLⅢon kidney in the TCM group and benazepril group were lower than the model group（ P〈0. 05）; the protein and gene expression of COLⅠ and COLⅢ on kidney of model group increased as time goes by,and the effect in the treatment group was still significant（ P〈0. 05）. Conclusion ＂Fushen Jiangzhuo Formula＂may reduce the accumulation of COLⅠ and COLⅢ on kidney to alleviate tubulointerstitial renal interstitial fibrosis and prevent the renal tubular interstitial damage by regulating the gene and protein expression of COLⅠ and COLⅢ on kidney.
