摘  要:目的研究桑葚花青素的体内外抗肿瘤作用。方法体内考察不同剂量的桑葚花青素对荷S180瘤小鼠体重抑瘤率及脾指数、胸腺指数和肝指数的影响。体外考察应用MTT法计算不同剂量的桑葚花青素对S180细胞株的生长抑制情况;采用流式细胞术检测桑葚花青素对S180细胞株的细胞周期与细胞凋亡的影响。结果 (1)不同浓度的桑葚花青素均有一定的抑制肿瘤的作用;桑葚花青素高剂量组与低剂量组比较,P<0.05;桑葚花青素高剂量组与阳性药组比较P<0.05。(2)阳性药物组和高剂量桑葚花青素组的脾指数及胸腺指数均明显高于空白对照组。(3)通过形态学观察,5-Fu阳性对照组和不同浓度桑葚花青素肿瘤细胞数量明显减少;且MTT实验表明,阳性药物组、低剂量组及高剂量组的肿瘤细胞生长抑制率分别为66.7%、54.2%及32.1%,阳性药物组与高剂量高于低剂量组(P<0.05)。(4)流式细胞术结果表明,桑葚花青素高剂量组、低剂量组及阳性药物组的肿瘤细胞处于S期的比例显著增高(P<0.05),而且各给药组细胞凋亡率也显著增高,与空白对照组相比,具有显著性差异(P<0.01)。结论桑葚花青素在体内外均有抗S180肉瘤的作用,值得更深入的研究。Objective To carry out preliminary study on in vivo and in vitro anti-cancer effects of anthocyanins in mulberry. Methods We observed effects of different doses of mulberry anthocyanins impact on S 180 tumor inhibition rate, spleen index, thymus index and liver index, meanwhile, we used MTT assay to calculate S 180 cell line growth inhibition. The changes of cell cycle and apoptosis of S180 were detected by flow cytometry. Results (I) Different concentrations of mulberry anthocyanins had effects of inhibiting tumor. (2) The spleen index and thymus index of positive drug group and high-dose group were significant higher than control group. (3) By morphological observation, number of tumor cells decreased in positive drug group mulberry andanthocyanins groups. MTT assay showed that the growth inhibition rate of tumor cells in positive drug group, low-dose group and high-dose groups, were 66.7%, 54.2% and 32.1% respectively. (4) Cell Cycle analysis indicated that anthocyanins in mulberry can arrest S 180 at S phase, Cell apoptosis analysis indicated that anthocyanins in mulberry can elevate the apoptosis rate,and they were dose-dependent manner. Conclusion Anthocyanins in mulberry has the role on anti-S 180 tumor in vivo and in vitro, we should carry out further study.
