摘  要:目的研究大孔树脂分离天仙子中总生物碱的最佳工艺条件。方法采用HPLC法测定,以东莨菪碱、莨菪碱的吸附量和解吸率为考察指标,分别考察东莨菪碱、莨菪碱在5种类型的大孔树脂(LSA5B、LSA21、LSD001、HPD600、D101)上的吸附和解吸附行为,并分析提取液p H、吸附体积流量、洗脱体积流量等参数对分离的影响。结果所比较的5种树脂中,LSA5B型大孔吸附树脂对天仙子总生物碱分离纯化效果最好,富集能力强,最佳富集工艺条件为以1.5 BV/h的体积流量上样,4 BV蒸馏水洗去水溶性杂质,最佳的洗脱溶剂为体积分数为50%乙醇,最佳洗脱体积流量为1.5 BV/h,洗脱用量为10 BV,东莨菪碱和莨菪碱的洗脱率可达74%和86%。结论经LSA5B型大孔吸附树脂纯化的天仙子总生物碱中东莨菪碱、莨菪碱可达1.85%、4.03%。AIM To optimize the processing conditions in the purification of the total alkaloids from Hyoscyami Semen by macroporous resin. METHODS The adsorption and desorption rates of scopolamine and hyoscyamine,as the indexes,were determined by HPLC. The best macroporous resin was screened out from five resins,LSA5 B,LSA21,LSD001,HPD600 and D101,and the parameters of p H value of loading solution,adsorption flow rate and desorption flow rate were also investigated. RESULTS Among the five macroporous resins,LSA5 B,with the best purification and enrichment potency for total alkaloids from Hyoscyami Semen,was considered the optimal one.The optimum enrichment process for alkaloids was so determined: at the loading flow rate of 1. 5 BV / h,wash off the water-solubility impurity with 4 BV water,and then enrich total alkaloids by 50% ethanol with a consumption of10 BV solution. The desorption efficiency of scopolamine and hyoscyamine could exceed 74% and 86%,respectively. CONCLUSION The content of scopolamine and hyoscyamine in total alkaloids prepared by LSA5 B can be1. 85% and 4. 03%,respectively.
