摘  要:目的研究达原饮对LPS诱导的大鼠发热模型的解热作用及其机制,采用UPLC/Q-TOF-MS法分析达原饮乙醇提取物中的化学成分。方法大鼠腹腔注射脂多糖(LPS)建立发热模型,药物处理后连续监测体温8 h,观察大鼠体温变化;运用酶联免疫试剂盒检测血清IL-6、TNF-α水平,MPO试剂盒检测肝组织MPO活性。采用UPLC/Q-TOF-MS法,分析达原饮乙醇提取物的化学成分。结果达原饮对LPS引起的大鼠发热具有明显的解热作用,显著降低血清中TNF-α、IL-6水平及肝组织中的MPO活性。正离子模式下检测出达原饮乙醇提取物中48个化合物。结论达原饮具有显著的解热作用,其作用机制与降低血清中IL-6、TNF-α水平,降低肝组织中MPO活性有关。通过UPLC/Q-TOFMS分析,达原饮的主要化学成分为黄酮、皂苷、生物碱和有机酸类化合物。AIM To observe the effect of Dayuanyin on LPS-induced febrile response in rats and elucidate the antipyretic mechanism of Dayuanyin,and to analyze the chemical composition in ethanol extract of Dayuanyin by UPLC / Q-TOF-MS. METHODS Fever was induced by intraperitoneally injecting LPS( 100 μg / kg),and the temperature was observed for 8 h,and the changes of temperature were observed. IL-6,TNF-α levels and liver tissue MPO levels in cell supernatant were detected. Chemical compositions of Dayuanyin were analyzed by UPLC / QTOF-MS. RESULTS Dayaunyin had significant antipyretic effect on rats induced by LPS,could reduce the level of TNF-α and IL-6 in serum,and decrease the activity of MPO in liver tissue. Forty-eight compounds in the ethanol extract of Dayuanyin had been detected in the positive ion mode. CONCLUSION Dayaunyin has significant antipyretic effect by decreasing the level of IL-6 and TNF-α in serum and reducing the content of MPO in liver tissue.Analyzed by UPLC / Q-TOF-MS,the main chemical compositions of Dayuanyin are flavonoids,saponins,alkaloids,organic acids.
