摘 要:目的建立同时定量测定狭基线纹香茶菜中viceninⅡ、isoschaftoside、schaftoside、vitexin的RH-HPLC。方法狭基线纹香茶菜70%乙醇提取液分析采用Phenomenex luna C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸水溶液梯度洗脱,体积流量0.8 m L/min,检测波长344 nm;柱温为室温(25℃)。结果 ViceninⅡ、isoschaftoside、schaftoside、vitexin线性范围分别为0.38~4.18μg、0.66~7.26μg、0.43~4.73μg、0.48~5.28μg;平均回收率(n=6)分别为97.7%、98.8%、96.9%、100.1%,RSD分别为1.6%、2.1%、1.8%、1.9%。结论本法的检测波长,流动相和提取溶剂的选择在狭基线纹香茶菜中黄酮碳苷类成分的定量分析上是最适合的。AIM To establish an RP-HPLC method for simultaneously determining the contents of viceninⅡ,isoschaftoside,schaftoside and vitexin from Isodon Lophanthoides. METHODS The analysis of ethanolic extrct of I. Lophanthoides was performed on Phenomenex luna C18( 250 mm × 4. 6 mm,5 μm) column and a mobile phase consisting of methanol and 0. 1% phosphoric acid in a gradient elution mode,flow velocity was 0. 8 m L / min and the detection wavelength was set at 344 nm,with the column temperature of 25 ℃. RESULTS The linear ranges of viceninⅡ,isoschaftoside,schaftoside,vitexin were 0. 38-4. 18 μg,0. 66-7. 26 μg,0. 43-4. 73 μg,and0. 48-5. 28 μg,respectively. Their average recoveries( n = 6) were 97. 7%( RSD = 1. 6%),98. 8%( RSD =2. 1%),96. 9%( RSD = 1. 8%),and 100. 1%( RSD = 1. 9%),respectively. CONCLUSION The method is the most suitable for quantitative determination of C-glycosyl flavones from I. Lophanthoides,especially in the selection of wavelength,mobile phase,and extract solution.
