摘 要:本实验通过使用1-甲基-3-硝基-1-亚硝基胍(NTG)诱变处理对数期嗜酸氧化亚铁硫杆菌(简称At.f菌),研究了同一时间段内与不同时间段内的不同浓度的诱变剂和不同诱变处理时间对诱变效应的影响,并在此基础上对诱变后菌株的多聚糖(简称EPS)变化率进行了探究。实验结果显示诱变后菌株的氧化活性在原始菌株的基础上提高约为5倍,诱变后的At.f菌的EPS变化率增加,反应了其氧化活性的增加。此外,通过诱变使得菌种的培养周期大大缩短。In the experiment, 1-methyl-3-nitro-1-nitrosoguanidine (NTG) was used for the mutagenic treatment of Acidthiobacillus ferrooxidans (At.f) at its logarithmic phase, and the influence of different concentrations of mutagen on the same time and different time intervals and different treatment time on the mutagenic effect were studied. In addition, tile rate of changes of the extracellular polysaccharides (EPS) on the modified strain was studied on the basis of the above. The experimental results show that the oxidation activity of the modified strains based on the original strain is increased about 5 times, and the rate of changes of EPS on the modified At.f is increased also indicated the increase of the oxidation activity of At.f. In addition, the training cycle of the strain is greatly shortened by mutation.
