摘  要:目的比较不同数量骨髓间充质干细胞治疗重型脑外伤的疗效。方法 48只成年SD大鼠随机分为3组治疗组和1组对照组,采用改良的Feeney氏法建立大鼠创伤性脑损伤模型,外伤后注射5×105(A组),2×106(B组),1×106(C组)BMSCs;采用行为测试试验评分观察肢体功能恢复,采用免疫组化法观察并比较4组大鼠制模后海马齿状回及损伤区域Brd U阳性细胞的表达。结果 A、B、C组前肢放置试验和平衡试验评分均优于对照组,C组优于A、B组(P<0.05)。A、B、C组中大鼠海马和损伤区域出现的Brd U阳性细胞数明显多于对照组,C多于A、B组(P<0.05)。结论三个治疗组均可促进肢体功能的恢复,但1×106治疗组治疗效果最为明显。Objective To compare the effect on different number of BMSCs treating heavy traumatic brain injury. Meth-ods 48 adult SD rats were divided into 3 treatment groups and the control group randomly, and the traumatic brain injury model established with improved Feeney’s method. 5 × 10 5 (A)、2 × 10 6 (B)、1 × 10 6 (C) BMSCs are given to rat at 24h after TBI respec-tively. The restoration of limbs function was observed by behavior test grading. BrdU-labeled cells of 4 groups were counted and compared in the dentate gyrus of the hippocampus and damage region by immunohistochemical method after TBI. Results Both of the grading of forelimb placing test and beam balance test examines in BMSCs transplantation with A、B、C group surpassed those of the control group, and that in C group better than with A、B group(P 〈 0. 05). Compare with the control group, more BrdU-labelde cells were detected in the dentate gyrus of the hippocampus and damage region after TBI in A、B、C group. And that in C group bet-ter than in A or B group(P 〈 0. 05). Conclusions Restoration of limbs function after TBI can be promoted by one of A、B、C group. However, this effect can be improved most significantly by 1 × 10 6 group.
