摘 要:目的研究去氢骆驼蓬碱在体外及小鼠体内抗细粒棘球蚴原头节的作用。方法通过体外药物干预试验检测去氢骆驼蓬碱(Harmine)体外杀原头蚴效果,并以去氢骆驼蓬碱对原头蚴的半数致死浓度(LC50)与传统抗包虫药物阿苯达唑(Albendazole,ABZ)相比较,评价去氢骆驼蓬碱体外抗细粒棘球蚴原头节活性;通过昆明白小鼠腹腔注射细粒棘球蚴原头节制备包虫病动物模型,观察10.0、5.0、2.5mg/kg体重去氢骆驼蓬碱连续给药14d对包虫病小鼠的囊湿重、抑囊率及囊泡和肝脏组织结构的影响。结果去氢骆驼蓬碱在体外可抑制原头蚴生长,LC50为(44.11±1.32)μg/ml;去氢骆驼蓬碱2.5、5.0、10.0 mg/kg体重均可抑制包虫病小鼠体内囊泡的生长,抑囊率分别为64.59%、68.38%和72.43%,且能破坏囊泡的生发层结构和抑制肝脏的炎症。结论去氢骆驼蓬碱对体内外细粒棘球原头蚴均有较强的抑制作用。Objective To study the anti-hydatid action of harmine on Echinococcus granulosus protoscoleces(EgPSC)by using harmine in vitro and in vivo. Methods Harmine was used as a drug intervention in vitro to determine the effectiveness with which it killed E.granulosus protoscoleces.The median lethal concentration(LC50)of harmine was compared with that of the conventional anti-hydatid drug albendazole(ABZ)to evaluate activity against E.granulosus protoscoleces in vitro.In addition,a mouse model was created by intraperitoneal injection of E.granulosus protoscoleces.Harmine was administered at concentrations of 2.5,5and 10mg/kg for 14 days to observe the effect of harmine on mean cyst weight and the rate of cyst inhibition.Histopathological changes in cysts were observed under both an optical microscope and an electron microscope,and histopathological changes in the liver were observed under an optical microscope. Results Harmine inhibited the growth of E.granulosus protoscoleces in vitro with an LC50of(44.11±1.32)μg/ml.Low,medium,and high doses of harmine significantly inhibited hydatidosis and cyst growth in mice.The rate of cyst inhibition by these doses was 64.59%,68.38%,and 72.43%,respectively.Doses damaged the layer structure of the hydatid and inhibited inflammation of the liver. Conclusion Harmine was highly effective at inhibiting E.granulosus cystsin vitro and in vivo.
