摘  要:【目的】研究蕨麻对模拟高原低压缺氧环境下大鼠脑组织保护作用及caspase 3表达的影响。【方法】78只健康成年雄性SD大鼠随机分为常氧对照组、低压缺氧组、蕨麻乙醇提取物高[3.6 g·(kg·d)-1]、中[1.8 g·(kg·d)-1]、低[0.9 g·(kg·d)-1]剂量组和尼莫地平[0.02 g·(kg·d)-1]组,每组13只。除常氧对照组外,其余各组大鼠均在减压舱内模拟海拔7 000 m缺氧12 h。测定脑含水量变化,HE染色观察大鼠脑皮质病理改变,透射电镜观察脑皮质神经元超微结构变化,RT-PCR检测caspase 3 m RNA表达,免疫组织化学和Western blotting方法检测caspase 3蛋白水平。【结果】与低压缺氧组比,蕨麻乙醇提取物[3.6 g·(kg·d)-1]组大鼠脑皮质形态结构接近常氧对照组,超微结构损伤显著减轻;脑组织的含水量显著降低;脑皮质caspase 3 m RNA及其蛋白的表达明显下调(P<0.01)。【结论】蕨麻乙醇提取物能够减轻低压缺氧导致的脑水肿,保护脑组织,此作用可能与其下调脑组织caspase 3表达有关。【Objective】To study the brain protective effect of Potentilla anserina L and its regulating on expression of caspase 3 in rats exposed to acute hypobaric hypoxia.【Methods】78 Male SD rats were randomly divided into control group, acute hypobaric hypoxia group, intervention groups of Potentilla anserina L. alcohol extract [3.6 g·(kg·d)^-1, 1.8 g·(kg·d)^-1, 0.9 g·(kg·d)^-1] and Nimodipine group[0.02 g·(kg·d)^-1], and each group with 13 rats. Each group with 13 rats. Pathological and ultrastructural changes in the cerebral cortex were observed respectively by HE staining and transmission electron microscope,the brain water content was determined. RT-PCR,Western blotting and immunohistochemistry assays were used for detecting the caspase 3 m RNA and protein.【Result】Compared with control group,the morphological strcture were improved and the ultrostructure injury were reduced in cortex of rats treated with 3.6 g·(kg·d)^-1Potentilla anserina L. alcohol extract. The brain water content reduced significantly. The levels of caspase 3 m RNA and protein were down-regulated obviously. 【Conclusion】Potentilla anserina L. alcohol extract could attenuate cerebral edema caused by hypobaric hypoxia and protect brain from injury. This activity may be related with its effect of down-regulating the expression of casepase 3.
