摘　　要：目的:制备受体相关蛋白(receptor-associated protein,RAP)单克隆抗体,研究人胰腺癌组织中RAP的表达与定位,探讨胰腺癌发生发展的病理机制。方法:运用基因重组技术及单克隆抗体技术制备抗RAP抗体。利用制备的抗RAP抗体,分别采用Western Blot和免疫组织化学检测并比较4例胰腺癌患者癌及癌旁组织中RAP的表达及分布。结果:抗RAP单克隆抗体能特异识别分子质量为44 ku的RAP蛋白。Western Blot与免疫组织化学检测结果均显示RAP在胰腺癌组织中的表达明显高于癌旁组织。免疫组织化学分析还显示RAP表达主要分布在细胞的胞质,呈强阳性;RAP在组织间质中也有少量分布,呈弱阳性。RAP在胰腺癌组织中的高表达主要分布在癌巢。结论:RAP表达与胰腺癌的发生发展密切相关。Objective： To Prepare the monoclonal antibody（m Ab） against receptor-associated protein（RAP）,study the expression and distribution of RAP in human pancreatic cancer,and to explore the pathological mechanism of the occurrence and development of pancreatic cancer. Methods： Gene recombination and monoclonal antibody preparation technology were used to prepare anti-RAP m Ab. The expression and distribution of RAP in the pancreatic cancer and the paracacinoma tissue of 4 patients after resection of the carcinoma of the panceas were detected respectively by Western Blot and immunohistochemistry by utilizing the prepared anti-RAP m Ab. Results： The anti-RAP monoclonal antibody could specifically identify the RAP protein with the molecular weight of 44 ku. Western Blot and immunohistochemistry both indicated that the expression of RAP in pancreatic cancer tissue was significantly higher than that in paracarcinoma tissue. Immunohistochemistry analysis also revealed that RAP was mainly expressed in cytoplasm of cells,showed strong positive. In interstitial tissue,RAP also had a small amount of the distribution,but showed weak positive. High expression of RAP in pancreatic cancer organizations are mainly distributed in the nests. Conclusion： The expression of RAP is closely related to the pathogenic progression of pancreatic cancer.
