摘  要:目的:探讨蛋白质组学技术用于动脉瘤性蛛网膜下腔出血(SAH)后脑血管痉挛(CVS)研究的可行性。方法:收集22例动脉瘤性SAH患者发病后第1天的脑脊液,按CVS程度将脑脊液标本分成3组:无痉挛组(对照组)、轻中度痉挛组、重度痉挛组。采用双向凝胶电泳分离各组混合脑脊液总蛋白,然后比较无痉挛组与轻中度痉挛组及重度痉挛组之间的双向凝胶电泳图谱。筛选出差异表达的蛋白质点,对差异点用4800 MALDI-TOF/TOF质谱仪进行鉴定。检索NCBInr数据库,确定差异表达的蛋白质。结果:无痉挛组与轻中度痉挛组及重度痉挛组差异表达2倍以上的蛋白质斑点有68个,成功鉴定并命名不同的差异表达蛋白23个。结论:运用蛋白质组学技术成功找到与CVS可能相关蛋白,为动脉瘤性SAH后CVS的发病机制的研究提供了新方法。Objective:To investigate the feasibility of proteomics applied in the study of cerebral vasospasm after aneurysmal subarachnoid hemorrhage (SAH).Methods:Cerebrospinal fluids of 22 patients after SAH within the first day after onset were collected.According to the degree of vasospasm,cerebrospinal fluid samples were divided into three groups:no vasospasm group (control group),mild-to-moderate vasospasm group,severe vasospasm group.Total protein of each group mixed cerebrospinal fluids was separated using two-dimensional gel electrophoresis,and then the two-dimensional gel electrophoresis map of no vasospasm group with mild-to-moderate vasospasm group and severe vasospasm group was compared.The differential protein spots were screened and the proteins were identified by the 4 800 MALDI TOF/TOF mass spectrometer,and then were determined according to the NCBInr databases.Results:We found 68 differential protein spots among mild-to-moderate vasospasm group,severe vasospasm group and no vasospasm group.There were 23 differentially expressed proteins which named differently successfully identified.Conclusion:We successfully found some proteins which may be correlated with cerebral vasospasm by using proteomics technology.It provides a new approach to study the pathogenesis of cerebral vasospasm after aneurysmal SAH.
