摘  要:目的观察康脑液对SD大鼠脑缺血再灌注后血管新生和碱性成纤维细胞生长因子(b FGF)、磷脂酰肌醇3激酶/蛋白激酶B(Pi3k/akt)表达的影响。方法雄性SD大鼠随机分为6组:3个剂量康脑液组[24,12,6g·(kg·d)-1]、尼莫地平组[1 mg·(kg·d)-1]、模型组及假手术组。栓线制备大鼠右侧大脑中动脉闭塞(MCAO)模型。于缺血2 h再灌注12,24,48,72,168 h,用免疫组化法观察b FGF、Pi3k及akt的表达。再灌注72 h用免疫组化法观察血管性血友病因子(v WF)的表达,计算微血管密度(MVD)。结果与模型组相比,康脑液组b FGF、Pi3k及akt表达明显增多,脑组织血管显著增多(P<0.05或P<0.01)。结论康脑液通过增加b FGF、Pi3k/akt的表达从而促进缺血区血管新生及减少缺血区神经功能损害,起到脑保护作用。Objective To observe the effect of Kangnao liquid ( KNY) on the expression of basic fibroblast growth factor ( bFGF ) , phosphatidyli-nositol 3-kinase/protein kinase B ( Pi3k/akt) and microvascular densi-ty ( MVD) in rats with focal cerebral ischemia -reperfusion.Methods The male SD rats were randomly divided into six groups:three dose KNY groups [ 24 , 12 , 6 g . ( kg . d ) -1 ] , nimodipine group [1 mg. (kg. d) -1], model group, sham group.The animal models of middle cerebral artery occlusion ( MCAO) were established by a monofi-lament method from right internal carotid artery.Immunohistochemical method was used to investigate the expression of bFGF and Pi 3 k/akt in rats on 12,24,48,72,168 h of reperfution injury after ischemia for 2 h and to mark neovascular vWF after 72 h of reperfution and to calculate MVD.Results Compared with model group , the expression of bFGF , Pi3k/akt at different time points and MVD of brain tissue were signifi-cantly increased of KNY groups ( P〈0.05 or P〈0.01 ).Conclusion KNY can increase the expression of bFGF and Pi 3 k/akt , promote tissue angiogenesis , then protect nerve cells of cerebral infarction areas.
