金复康口服液抑制Lewis肺癌Sca-1^+干细胞亚群增殖的研究

摘　　要：目的观察金复康口服液抑制肺癌干细胞增殖和成瘤性作用及其对耐药相关蛋白表达的影响。方法采用免疫磁珠法分选Lewis肺癌Sca-1+细胞,体外采用金复康含药血清进行干预,综合应用流式细胞术、免疫组化、免疫印迹法、RT-PCR等实验技术观察金复康口服液抑制Sca-1+干细胞亚群增殖的作用,以及逆转肺癌干细胞耐药性基因ABCG2和蛋白表达的作用。结果体外研究发现,金复康口服液可时间依赖性地抑制肺癌Sca-1+干细胞在体外的增殖(P<0.05);诱导肺癌Sca-1+干细胞发生凋亡;金复康口服液单独应用或者与化疗联合均可有效下调ABCG2基因的转录和蛋白的表达(P<0.05,P<0.01)。结论金复康口服液可显著抑制Lewis肺癌Sca-1+干细胞亚群增殖,其机制可能和抑制耐药相关蛋白的表达有关。Objective To evaluate the inhibitory effect of ＂Jinfukang Decoction＂ on lung cancer stem cell proliferation and tumorigenesis, and to explore its effect on multidrug related protein expression regulation. Methods lmmunomagnetic beads was applied to sorting the Sca-1^＋ Lewis lung cancer cell. In vitro study conducted to assess the inhibitory of the cell proliferation by MTT, with a serum containing the ingredients of ＂Jinfukang Decoction＂ as the intervention ; Western-Blotting and RT-PCR were used to observe the effect of reversing the expression of muhidrug resistance ABCG2. Results In vitro study founded that ＂Jinfukang Decoction＂ was able to inhibit the proliferation of the cell with time-dependent manner, and was also able to induce the apoptosis of the Sca-1^＋ Lewis lung cancer cell, comparing with a control group （ P 〈 0.05 ）. ＂Jinfukang Decoction＂ alone or combined with chemotherapy could down-regulate the transcription and expression of the muhidrug resistance gene remarkblely, comparing with a control group （P 〈 0. 05 ）. Conclusion ＂Jinfukang Decoction＂ can significantly inhibit the proliferation f the Sea-1 ^＋ Lewis lung cancer cell by decreasing the expression of muhidrug resistance protein ABCG2.
