摘 要:滞育是昆虫逃避不利环境的基本方式之一,但目前仅极少数基因被证明直接参与了昆虫滞育的调控。综述了昆虫滞育关联差异基因的筛选方法及相关方法的应用概况,包括差异显示反转录PCR、构建全长c DNA文库、基因表达序列标签、基因芯片技术和RNA转录组测序等在滞育差异表达基因筛选中的研究应用。对蛋白质组学和RNA干扰技术在昆虫滞育调控中关联蛋白和基因的功能研究一并进行了总结。为昆虫滞育调控机制中关键基因的发掘、滞育进程中对逆境条件耐受机制和探索害虫生态调控的新途径提供参考。Diapause is one of the basicmodes for insects to escape from unfavorable environment,however, there are very few genes which are proved to be involved in the regulation of diapause.This paper summarizes the methods for screening differential expression genes related to insect diapause including display reverse transcription PCR,construction of full length cDNA library,expressed sequence tags,microarray and RNA-Sequencing technology,which have been used in screening differentially expressed genes related to insect diapause.The proteomics and RNA interference technology used to exploring the function of key proteins and genes associated with the regulation of insect diapause are also summarized.This review may provide some valuable reference for exploring key genes in insect diapause regulating mechanism,understanding the mechanisms of diapausing insect response to reverse environmental conditions and exploring new ways for ecological pest management.
