摘  要:目的 初步探讨层流对血管内皮的保护机制.方法 将牛主动脉内皮细胞在特殊材料的装片上培养并置于特殊的容器内,由计算机发出的信号分别传给泵/控制板和容器,用于控制来自装瓶溶液的流速/流动方向.内皮细胞被机械力作用后,再收集细胞进行检测.结果 二氢叶酸还原酶(DHFR)在层流(15 dyns/cm^2)作用下表达上调,同时伴有氧自由基的下调和一氧化氮的合成增加,细胞间黏附因子(ICAM-1)的表达下降.转染DHFR RNA干扰后,上述变化消失.结论 DHFR的上调可能介导了层流对内皮细胞功能的保护作用.Objective To investigate the possible protective mechanism of laminar shear on en- dothelial cells fuction. Methods The bovine aortic endothelial cells cultured on slides were placed in a special container. The signal sent by the computer was passed to the pump/control panel and the container which was used to control the flow rate and direction of the solution from the reservior. Endothelial cells were exposed to mechanical force which were collected afterwards for measurements. Results Dihydrofo- late reductase( DHFR)was upregnlated by laminar flow( 15 dyns/cm^2) which accompanied by increased production of nitric oxide, decreased superoxide and downregnlation of intercellular adhesion molecule-1 ( ICAM-1 ). However, the above changes were resumed after DHFR RNAi transfection. Conclusion DH- FR may mediate the protective effect of laminar flow on endothelial cells function.
