摘  要:目的探讨咪喹莫特诱导的银屑病样Balb/c小鼠模型及其皮肤血管增生的特点。方法备皮后,以外涂凡士林为对照组,在Balb/c小鼠背部皮肤外涂5%咪喹莫特乳膏为模型组,连续6d,诱导产生银屑病样皮损。用常规病理检查观察模型构建情况;用免疫组化染色方法,分别对模型组与对照组皮损组织进行CD31染色,以计算皮损组织内微血管密度,并观察微血管形态。结果常规病理结果提示,5%咪喹莫特乳膏能诱导Balb/C小鼠产生银屑病样皮损,表现为表皮角化异常、炎症细胞浸润、血管增生等银屑病病理特点;而对照组无改变。免疫组化结果显示,对照组及模型组均有不同程度的CD31表达,主要表达在微血管内皮细胞上。通过Weidner法计算小鼠皮损的微血管密度,提示模型组小鼠皮损的微血管密度(17.11±2.36)个/mm2较正常对照组(3.18±1.14)个/mm2明显增大(t=15.030,P<0.001)。模型组微血管在真皮浅中、层增多明显,且管腔形态迂曲扩张。结论 5%咪喹莫特乳膏可以诱导Balb/c小鼠产生银屑病样皮损,且该皮损具有与人类银屑病相似的血管增生特点。Objective To evaluate the angiogenesis of imiquimod-induced psoriatic mouse(Balb/c) model. Methods Topical 5%imiquimod cream was used for 6 consecutive days on the back skin of Balb/c mice to induce psoriastic skin lesions. The mice treated with vaseline were taken as control. H&E staining was used to evaluate the modeling effect. The skin lesions were also stained by CD31 with immunohistochemical staining, and microvessel density and its characteristics in dermis were studied. Result Topical imiquimod cream could induce psoriatic lesions in Balb/c mice sharing most of histological features of psoriasis, including inordinate keratinisation, cutaneous inflammatory and angiogenesis conditions. No cutaneous change was found in the control group. Stained with CD31,the immunohistoehemieal results showed that microvessel density in the model group (17.11 -2.36) was significandy higher than that of the control (3.18±1.14, t =15.030, P 〈0.001). The proliferating microvessels of model group mostly distributed in superficial and middle layers of dermis, showing circuitous and expansing. Conclusion Topical 5% imiquimod cream Can induce psoriasitie features on Balb/c mice with hyperlasia of anglogenesis mimicking the features of human psoriasis.
