摘  要:本研究克隆了水稻白叶枯病病原(Xanthomonas oryzae pv.oryzae,Xoo)菌株JXOV的2个avrBs3/pthA家族成员基因,分别命名为jva1和jva2。通过氨基酸序列分析和系统发育分析、原核表达载体构建及在水稻品种叶片接种试验,探讨其基因功能和毒性。结果表明,Jva1和Jva2的氨基酸序列具有Avr Bs3/pth A家族成员典型序列模式特征;BL21(DE3)原核表达的Jva1和Jva2蛋白在SDS-PAGE胶上呈现出推测的100KD单体蛋白大小,没有出现不依赖于植物蛋白的Avr Bs3类似的二聚体化;jva1和jva2没有增强PXO99A在携带不同抗性基因的15个水稻品种上的毒性。jva1和jva2的基因功能有待于进一步研究。本研究结果为诠释水稻白叶枯病原的致病机理及相关基因家族成员的功能分化提供了重要参考。Two members of avrBs3/pthA family were cloned from Xanthomonas oryzae pv. oryzae(Xoo) race JXOV strain,and named as jva1 and jva2,respectively. We conducted the amino acid sequence analyses and phylogenetic analyses and constructed the prokaryotic expression vectors to investigate the gene function and toxicity of jva1 and jva2. The results showed that the deduced amino acid sequences of Jva1 and Jva2 shared typical characteristics with other gene members of Avr Bs3/pth A family. The molecular mass of proteins jva1 and jva2 from BL21(DE3) is about 100 KD on SDS-PAGE which is similar to presumed monomer protein. Dimerization of jva1 or jva2 did not occur in prokaryotic cell BL21,although the dimerization of Avr Bs3 was independent of plant proteins; Neither jva1 nor jva2 enhanced the PXO99^A strain virulence on fifteen rice cultivars holding different resistance genes,The gene functions of jva1 and jva2 need further investigation. These results provided important basis for understanding mechanism of pathogenicity of Xoo on rice and gene functionary diversity of members of related gene family.
