摘  要:目的探讨妊娠期糖尿病(GDM)胎盘组织细胞中性激素结合球蛋白(SHBG)和葡萄糖转运蛋白(GLUT1、GLUT3、GLUT4)的表达及相关性。方法应用免疫细胞化学染色法检测SHBG与GLUT1、GLUT3、GLUT4在30例GDM孕妇和30例正常孕妇胎盘组织中的表达情况。结果在GDM孕妇胎盘组织中,SHBG、GLUT1、GLUT3、GLUT4阳性细胞的平均光密度值分别为0.401 6±0.014 8,0.405 8±0.012 3,0.402 0±0.012 1,0.391 3±0.008 4。在正常孕妇胎盘组织中,阳性细胞的平均光密度值分别为0.414 2±0.011 5,0.427 0±0.029 4,0.418 0±0.008 7,0.402 9±0.095 9。比较SHBG与GLUT1、GLUT3、GLUT4在GDM孕妇和正常孕妇胎盘组织中的表达水平,差异均有统计学意义(P<0.05)。Pearson相关分析结果显示:SHBG和GLUT4在胎盘组织中的表达量呈正相关(R2=0.410 4,P<0.05)。结论 GLUT1是胎盘中最主要的葡萄糖转运载体,可能在母婴间葡萄糖转运中发挥主要的作用。GLUT4在胎盘组织表达较少,却受SHBG表达的影响。Objective To investigate the expression of SHBG and GLUT proteins in the placenta tissue of gestational diabetes mellitus ( GDM ), and analyze their correlation. Methods Expression of SHBG, GLUT1, GLUT3, and GLUT4 were detected by immunocytochemical staining in pla- centa tissues from 30 cases of GDM pregnant women and 30 cases of normal pregnant women. Results The average optical density value of positive cells of SHBG, GLUT1, GLUT3, and GLUT4 in the placenta tissue of GDM pregnant women were 0.401 6 ±0.014 8,0.401 6±0.012 3,0.402 0± 0.012 1, and 0.391 3±0.008 4, respectively, which were 0.414 2±0.414 2,0.427 0±0.029 4,0.418 0±0.008 7, and 0.402 9±0.095 9 in the normal pregnant . The SHBG and GLUT protein expression in GDM pregnant women and normal women organizations were statistically different (P 〈 0.05 ). Results of Pearson correlation analysis show that SHBG and GLUT4 expression in placenta tissue were positively correlated (R2--0.410 4, P 〈 0.05). Conclusion GLUT1 is the most important glucose transporter in the placenta, and plays the main role in glucose transport between mother and baby. The GLUT 4 mainly plays an important role in the insulin pathway, and SHBG was involved in the regulation of the insulin pathway.
