摘 要:目的探讨低甲状腺素血症大鼠是否存在胰岛素抵抗(IR)及其可能机制。方法将12只SD大鼠随机分为正常对照(NC)组及低甲状腺素血症(HypoT4)组。NC组正常喂养,HypoT4组给予丙硫氧嘧啶(PTU)水(浓度为3ppm)喂养。喂养8周后,眶后采血测定促甲状腺素(TSH)、总甲状腺素(TT4),用高胰岛素—正葡萄糖钳夹技术评估2组大鼠的胰岛素敏感性。应用ELISA方法检测大鼠血清胰岛素水平,通过Real-time PCR检测脂肪细胞的瘦素(Ob)和脂联素(APN)mRNA的表达水平。结果HypoT4组大鼠的血清TSH水平与NC组相比无统计学差异(P>0.05),但TT4明显低于NC组(P<0.05),2组大鼠的基础状态血糖和稳态血糖无统计学差异(P>0.05);HypoT4组的基础胰岛素水平有下降趋势,但较NC组无统计学差异(P>0.05),钳夹稳态状态下胰岛素水平明显高于NC组(P<0.05);HypoT4组的葡萄糖清除率和葡萄糖代谢率明显低于NC组(P<0.05),HypoT4组Ob mRNA表达水平低于NC组(P<0.05),APN mRNA表达水平低于NC组,虽差异无统计学意义,但有明显下降趋势(P=0.07)。结论低甲状腺素血症大鼠存在胰岛素抵抗,并可能与脂肪细胞分泌Ob及APN减少有关。Objective To investigate whether the insulin resistance (IR) exists in the rats with propylthiouracil (P]3J)-induced hypothyroxinemia and explore its possible mechanism. Methods A total of 12 SD rats were randomly divided into two groups : normal control group (NC) and hypo- thyroxinemia group (HypoT4). The NC group was normally fed, and the HypoT4 group was fed with PTU water (3 ppm). Eight weeks later, the blood samples of both groups were collected to measure the TSH and TF4. The hyperinsulinemic-euglycemic clamp technique was used to assess the insulin sensitivity and the serum insulin level was determined by EL1SA method. Real-time PCR technology was used to measure the expression level of Leptin mRNA and Adiponeetin mRNA in adipocytes. Results There was no significant difference in TSH level between two groups (P 〉 0.05 ), but TI'4 of the HypoT4 group was obviously lower than that of NC group (P 〈 0.05). No statistical difference was found between two groups about the basic blood glucose (BBG) and the homeostasis blood glucose (HBG) (P 〉 0.05 ). No statistical difference was observed between two groups on the basic insulin (Bins) (P 〉 0.05 ), but the insulin level was tending downwards in the Hypo% group. Under the clamp steady state, the insulin level of the Hypo% group was obviously higher than the NC group (P 〈 0.05 ). The glucose disposal rate (GDR) and the glucose metabolic rate (GMR) of the Hypo% group were lower than that oftbe NC group (P 〈 0.05). The Leptin mRNA expression level of the HypoT4 group was lower than the NC group (P 〈 0.05 ). The Ach'ponectin mRNA expression level was lower than the NC group with a clear declining trend, however, there is no statistical difference (P = 0.07 ). Conclusion Insulin resistance exists in the rats with hypothyroxinemia, which is possibly relevant to the re- dueed excretion of Leptin and Adiponectin in adipocytes.
