摘  要:目的探讨川芎嗪对大鼠脊髓损伤(SCI)后运动功能恢复的影响及可能的机制。方法健康成年Sprague-Dawley大鼠60只,随机分为川芎嗪治疗组(TMP组)、单纯脊髓损伤组(SCI组)和假手术组(Sham组),每组20只。Sham组只暴露脊髓,不打击。SCI组和TMP组采用Allen法损伤大鼠T9脊髓节段,复制SCI动物模型。造模成功后30 min,TMP组腹腔注射川芎嗪200mg/kg,每日1次,共7次;SCI组和Sham组腹腔注射等体积的生理盐水。于术后1、3、7、14 d随机抽取各组动物5只,进行后肢功能Basso-Beattie-Bresnahan(BBB)评分、脊髓组织HE染色、脊髓组织中白细胞介素(IL)17和磷酸化的信号转导与转录激活因子3(p STAT3)表达水平的检测。结果与Sham组相比,SCI组术后各时间点BBB评分均显著降低,形态学出现典型的SCI改变,脊髓组织中IL-17和p STAT3表达水平显著升高(P<0.05)。与SCI组相比,TMP组术后3、7、14 d BBB评分均显著升高,SCI改变减轻,脊髓组织中IL-17和p STAT3表达水平显著降低(P<0.05)。结论川芎嗪可通过抑制STAT3活性降低IL-17水平,促进SCI大鼠运动功能的恢复。Objective To observe the effect and mechanism of tetmmethylpyrazine (TMP) on motor function 'after spinal cord injury ( SCI ) in rats. Methods Totally 60 healthy adult Sprague-Dawley rats were divided randomly into TMP group, Sham group and SCI group with 20 rats in each group. The Sham group was only exposed spinal cord without imposing any harm. SCI group and TMP group were treated with Allen' s mode at the level of the 9th thoracic vertebra to imitate spinal cord injured animal models. 30 min "after SCI model was made, TMP group was given 200 mg/kg TMP daily for 7 times by intraperitoneal injection, while SCI group and Sham group were given an equal volume of nornml saline daily. On the 1 st, 3rd, 7th and 14th days after SCI, 5 rats were selected randomly from each group. The motor function was assessed by Basso-Beattie-Bresnahan (BBB) scale. The pathological changes of spinal cord tissue were determined by HE staining. The interleukin 17 (IL- 17 ) and phosphorylated signal transducer and activator of transcription 3 ( pSTAT3 ) expression were determined by Western blot. Results Compared to Sham group, the BBB scores in SCI group were significantly increased, HE staining presented typical pathological changes of spinal cord injury in SCI group, meanwhile the expression of IL- 17 and pSTXI3 in the spinal cord tissue in SCI group were all significantly increased (P 〈 0.05 ). Compared to SCI group, BBB scores in TMP group were significantly increased on the 3rd, 7th and 14th days after SCI, the pathological changes of spinal cord injm'y in TMP group was alleviated on the 3rd, 7th and 14th days after SCI, the expression of IL- 17 and pSTAT3 in TMP group were reduced on the 3rd, 7th and 14th days (P 〈 0.05 ). Conclusion TMP may reduce the expression of IL- 17 by inhibiting the activity of STAT3 and pl^mote the recovery of the motor function in rats after SCI.
