线性免疫印迹法检测抗60 kD Ro/SSA IgG抗体的对比分析

摘　　要：目的评价5个不同厂家自身抗体谱线性免疫印迹法(LIA)试剂中的抗60 k D Ro/SSA Ig G抗体项目的一致性及符合率。方法收集本院54例自身免疫性疾病病人和10份2013年卫生部临检中心室间质评血清及30例健康人血清,同时使用酶联免疫吸附(ELISA)法和5个不同品牌LIA法试剂对血清中的抗60 k D Ro/SSA Ig G抗体进行检测,比较5个不同品牌LIA法试剂检测60 k D Ro/SSA Ig G抗体的一致性及总体符合率。结果 ELISA法试剂盒检测94例血清中阳性标本37例,阴性标本57例,同时进行的5种不同品牌LIA法抗核抗体谱试剂检测抗60 k D Ro/SSA Ig G抗体结果的总体符合率均超过90%,吻合系数均>0.8,说明一致性很好,但5种品牌均出现不相一致的阳性和阴性。结论 LIA法的抗核抗体谱检测试剂盒在检测抗60 k D Ro/SSA Ig G抗体项目中一致性很好,但检测结果可疑时应使用天然抗原包被的ELISA法试剂或双向扩散法进行复测。Objective To evaluate the accordance and coincidence rate of five different autoantibody linear immunoassays（ LIA） for measurement of serum Ig G antibody against 60 k D Ro/SSA Ig G. Methods Fifty four serum samples from clinically confirmed autoimmune disease patients,10 from National Center for Clinical Laboratory originally distributed for external quality assessment in 2013 and 30 from healthy volunteers were measured by ELISA Kits and five different autoantibody LIA for 60 k D Ro / SSA Ig G and their accordance rates were evaluated. Results Among 94 samples measured by ELISA Kits,37 were positive while 57 were negative. The accordance rates among five different autoantibody LIA for 60 k D Ro / SSA Ig G were all over 90%,with all matching coefficients more than 0. 8. These results show that the accordance rates of five different autoantibody LIA are very good. However,different levels of positivity and negativity are also found among them. Conclusion Five different autoantibody LIA are all reliable for measuring serum anti-Ro / SSA Ig G antibody,but there are different levels of positivity and negativity. So if the test result is doubtful,ELISA or double diffusion test should be used to validate the result.
