摘 要:目的探讨桔梗中皂苷类组分(SCPG)对肉瘤S180鸡胚绒毛尿囊膜(CAM)移植瘤血管生成的影响。方法建立肉瘤S180 CAM移植瘤模型,并将模型样本随机分为五组:模型对照组,阳性药参一胶囊组(50μg·m L-1),低剂量SCPG组(25μg·m L-1),中剂量SCPG组(50μg·m L-1)和高剂量SCPG组(100μg·m L-1)。给药6 d后观察移植瘤形态,测定移植瘤体积、瘤重及抑瘤率,并运用免疫组化SABC法对移植瘤组织微血管密度(MVD)和血管内皮生长因子(VEGF)进行测定。结果 SCPG 25μg·m L-1、50μg·m L-1、100μg·m L-1三个剂量组对肉瘤S180 CAM移植瘤均有不同程度的抑制作用,其抑瘤率分别为25.70%、36.93%、48.18%,与模型组比较有显著差异(均P<0.05);检测各剂量组移植瘤MVD均值分别为(18.6±1.5),(18.2±1.3)和(16.2±1.3),与模型组比较有显著差异(均P<0.05);VEGF检测显示各剂量组均有不同棕黄色阳性表达,随剂量的增大,其着色程度呈逐渐减弱的趋势。结论 SCPG抑制肉瘤S180 CAM移植瘤生长及血管新生,其作用机制可能是通过抑制VEGF的表达,从而影响肿瘤生长因子与抑制因子之间的平衡而起到抑制肿瘤的作用。AIM To investigate the effect of saponin components from Platycodon on the angiogenesis in transplanted tumor of S180 sarcoma of chick chorioallantoic grandiflorum membrane (SCPG) CAM) .METHODS To establish S180 sarcoma model of CAM, and the model were randomly divided into 5 groups, control group, Shen Yi capsule group (50 μg·mL^-1 , low (25 μg·mL^-1) , middle (50μg·mL^-1) and high (100μg·mL^-1) dose SCPG groups, respectively. After 6 days of intervention, the transplantation tumor morphology, tumor size, tumor inhibitory rate were measured, and the samples were evaluated with SABC immunohistochemical reaction to examine the vascular endothelial growth factor (VEGF) expression and microvessel density (MVD) count. RESULTS Three dose groups of SCPG have activities of inhibiting the growth of S180 sarcoma implanted on the CAM, the mean inhibition rates were 25.7%, 36.93% and 48.18% (P 〈 0.05), respectively. MVD count was (18.6 ± 1.5), (18.2 ± 1.3), and (16.2 ± 1.3) respectively in all SCPG groups (P 〈 0.05). Immunohistochemical assay showed that the VEGF of the treated groups have some inverse proportion with the effect of anti-angiogenesis and anti-tumor. CONCLUSION SCPG have activities of inhibiting the growth of S180 sarcoma implanted on the CAM and inhibits the angiogenesis. Its anti- tumor mechanism is related with reducing the expression of VEGF to regulate the balance between tumor growth factors and inhibitors.
