摘 要:目的探讨盆腔浆液性肿瘤干样细胞(cancer stemlike cells,CSC)比例与腹腔转移的关系。方法 2011年7月至2013年7月共采集17例未行化疗的女性盆腔浆液性肿瘤患者的肿瘤原发灶及腹水标本,分别采用Hoechst33342拒染法检测侧群(side population,SP)细胞、乙醛脱氢酶(aldehyde dehydrogenase,ALDH)标记法检测ALDH+细胞,观察两者间SP细胞及ALDH+细胞的表达比例差异,并分析其与盆腹腔转移之间的关系。结果在可检测的标本中,73.3%(11/15)的患者肿瘤原发灶或腹水中SP细胞检测阳性(SP细胞比例>0.15%),100%(10/10)的患者原发灶或腹水中检测到ALDH+细胞。比较患者不同部位SP细胞和ALDH+细胞比例,可见73.3%(11/15)的患者腹水中SP细胞明显高于原发灶(P=0.01),90%(9/10)的患者腹水中的ALDH+细胞比例高于原发灶(P=0.03),且腹水与原发灶中SP细胞及ALDH+细胞均高的患者与仅SP细胞或ALDH+细胞高的患者发生肿瘤盆腹腔转移的比例差异无统计学意义(P>0.05),但其盆腹腔转移部位数明显较后者多(P<0.05)。结论盆腔浆液性肿瘤患者腹水中CSC比例明显高于原发灶,推测CSC与肿瘤盆腔种植转移和进展有关。Objective To investigate the relationship between pelvic serous cancer stemlike cells (CSC) and peritoneal metastasis. Methods Primary pelvic serous cancer tissues and aseites were obtained from 17 female patients without preoperative chemotherapy from July 2011 to July 2013. Side population (SP) cells and aldehyde dehydrogenase positive (AI.DH~) cells were tested by Hoeehst33342 staining and ALDH staining respectively to observe their expression difference and their relationship with pelvic metastasis. Results Among the detectable specimen,73.3% (11/15) were detected SP cells positive (percentage of SP cells〉0.15 % ), and 100 % (10/10) ALDH ^+cells in either primary foci or ascites. Comparing the cells percentage from different sites in the same patients,73.3 % (11/15) of the patients showed higher SP cells in ascites than those in primary tumors (P = 0.01 ) , whereas 9(1% (9/10) of the patiens showed higher ALDH+ cells in ascites than those in primary tumors (P = 003). Furthermore, patients with both SP positive and more ALDH+ cells in aseites showed more metastatic sites than those with only SP positive or AI.I)H+ cells(P〈0.05), however, they had little difference in metaststic rate (P〉0. 05). Conclusions Higher percentage of SP cells and ALDH ^+ cells in ascites indicates that pelvic serous CSC are related with peritoneal metastasis and disease progression.
