摘  要:目的:建立胰岛素抵抗大鼠模型,探讨血清网膜素-1水平与胰岛素抵抗的相关性。方法:30只SPF级雄性Wistar大鼠随机分为正常对照组(NC,n=15)、高脂饮食组(HF,n=15),自由摄食、饮水,NC组给予普通饲料,HF组给予高脂饲料诱导胰岛素抵抗模型。10周后2组各取5只行高胰岛素正葡萄糖钳夹实验评估模型,ELISA法检测血清网膜素-1,放射免疫法检测血清基础胰岛素。结果:(1)各组大鼠基础血糖比较无显著差异(P>0.05)。(2)HF组血清基础胰岛素水平较NC组明显升高(P<0.05),但血清网膜素-1水平较NC组明显降低(P<0.01)。(3)Pearson相关性分析示血清网膜素-1分别与胰岛素(r=-0.654,P<0.01)和游离脂肪酸(r=-0.446,P<0.05)呈负相关。结论:在大鼠胰岛素抵抗阶段,血清网膜素-1水平已经开始降低;基础胰岛素水平随着血清网膜素-1水平的降低而升高,推测血清网膜素-1水平降低可能成为胰岛素抵抗、糖尿病及心血管疾病的早期提示因子。AIM: To establish the insulin resistance rat model for evaluating the correlation of omentin-1 level and insulin resistance. METHODS : SPF male Wistar rats ( n = 30) were randomly divided into normal control group (NC, n = 15) and high-fat diet group (HF, n = 15). The rats in NC group were fed with basic diet. The insulin resistant model was established by feeding the rats with high-fat diet in HF group. After 10 weeks, 5 rats in each group were as- sessed by the technique of hyperinsulinaemic-euglycaemic clamp. After the insulin resistant model was successfully estab- lished, the body weight and fasting blood glucose were detected. The concentration of fasting serum omentin-1 was analyzed by ELISA. Fasting serum insulin was measured by radioimmunoassay. RESULTS: No difference of fasting blood glucose between the 2 groups was observed. The level of fasting serum insulin in HF group was significantly higher than that in NC group (P 〈 0. 05 ). The level of serum omentin-1 in HF group were significantly decreased compared with NC group (P 〈 0. 01 ). Pearson's correlation analysis showed that negative correlations between serum omentin-1 and fasting serum insulin ( r = - 0. 654, P 〈 0. 01 ), serum omentin-1 and free fatty acid ( r = - 0. 446, P 〈 0. 05 ) was found. CONCLU- SION: In rats, serum omentin-1 level began to decrease at insulin resistance stage. As serum omentin-1 level decreased, the basal insulin level increased, indicating that decreased serum omentin-1 level may be an early factor of IR, diabetes and cardiovascular diseases.
