摘 要:目的探讨不同粒径的纳米二氧化硅(nm-Si O2)与微米二氧化硅(μm-Si O2)对人肺腺癌上皮A549细胞(A549细胞)的细胞毒性及其炎性因子分泌的影响。方法选择3种粒径的纳米Si O2(20、60、100 nm)和微米Si O2(0.1-5μm)按照不同作用浓度(0、12.5、25、50、100、200μg/m L)对A549细胞进行染毒,24 h后采用四甲基偶氮唑盐微量酶反应比色(MTT)法测定A549细胞的存活率,采用微量酶标法测定乳酸脱氢酶(LDH)活性、白细胞介素8(IL-8)和肿瘤坏死因子(TNF-α)含量。结果 3种粒径nm-Si O2均可引起细胞存活率降低,细胞释放LDH升高,存在剂量效应关系;20-nm-Si O2对细胞的毒性作用最大,在较低剂量25μg/m L时可使细胞存活率显著低于对照组(P〈0.01),在染毒浓度相同的条件下,20-nm-Si O2组LDH活力显著高于60-nm-Si O2组(P〈0.01)、60-nm-Si O2组LDH活力显著高于100-nm-Si O2组(P〈0.05);μmSi O2仅在最高剂量时引起细胞存活率降低(P〈0.05)。3种粒径nm-Si O2均可使细胞分泌IL-8的量增加,但是并未引起TNF-α变化,在染毒浓度为50μg/m L时,20-nm-Si O2组IL-8的分泌量均显著高于100-nmSi O2组(P〈0.01)。μm-Si O2未引起细胞分泌IL-8和TNF-α的变化。结论 3种粒径nm-Si O2均可引起A549细胞的细胞毒性,引起细胞炎性因子IL-8分泌增加,且nm-Si O2粒径越小,其毒性效应愈强,但并未引起TNF-α变化;μm-Si O2对A549细胞的毒性效应不明显。Objectives To study the effects of nano-silica (nm-SiO2) and micro-silica (μm-SiO2) particles on the cytotoxicity and secretion of inflammatory factors in human lung adenocarcinoma cells (A549 cells). Methods A549 cells were exposed to three sizes of nm-SiO2(20, 60 and 100 nm) and μm-SiO2(0. 1 -5 μm) particles for 24 h at different doses ( 0, 12.5, 25, 50, 100 and 200 μg/mL). The cytotoxicity of nm-SiO2and μm-SiO2 on A549 cells was measured by MTT method, the activity of LDH and the levels of IL-8 and TNF-α were measured by micro-ELISA method. Results The viability of cells was inhibited and the activity of LDH was increased with a dose-response relationship in all nm-SiO2 groups. The cytotoxicity of ceils in 20 um-SiO2 group was most serious, even in a lower dose of 25 μg/mL; the viability of cells in 20 nm-SiO2 group was significantly lower than that of control group (P 〈 0.01 ). The activity of LDH in 20 nm-SiO2 group was significantly higher than that of 60-nm-SiO2 group, and the activity of LDH in 60 nm-SiO2 group was higher than that of 100-nm-SiO2 group (P 〈 0.05) ; but the viability of cells in μm-SiO2 groups was reduced only in the highest dose group (P 〈 0.05 or P 〈 0.01 ). The secretion of IL-8 in cells was increased in 3 nm-SiO2 groups, but no significant changes in the secretion of TNF-α from cells, the secretion of IL-8 from cells in 20-nm-SiO2 group was significantly higher than that in 100-nm-SiO2 group (P 〈0.01). However, the secretion of IL-8 and TNF-α was not increased significantly in μm-SiO2 groups. Conclusions Three nanometer sized SiO2 particles could cause cytotoxicity and increase the secretion of IL-8 in A549 cells; and the smaller size of nm-SiO2particles were, the stronger of its toxic effects; but no significant change was observed in the secretion of TNF-α in the cell in nm-SiO2groups. However, μm-SiO2 parti- cles could not cause significant cytotoxicity in A549 cells.
