摘 要:研究旨在检测血红素加氧酶(HO-1)和胆绿素还原酶A(BLVRA)在卢氏鸡和固始鸡输卵管子宫部的表达量差异,探索绿壳蛋的形成机理。以24周龄产绿壳蛋卢氏鸡和产褐壳蛋固始鸡为素材,采集每只鸡输卵管子宫部,提取总RNA,经过RT-PCR反转录为c DNA,以与色素相关的血红素加氧酶-1(Heme Oxygenase,HO-1)和胆绿素还原酶A(BLVRA)为目的基因,选择GAPDH作为参照基因,通过实时荧光定量PCR,分析HO-1和BLVRA在不同品种鸡的机体内转录水平的差异。结果表明:卢氏鸡子宫部组织中HO-1的m RNA相对表达量显著高于固始鸡(P<0.05)。固始鸡子宫部组织中BLVRA m RNA相对表达量极显著高于卢氏鸡(P<0.01)。This study aims to detect the expression difference of HO- 1 and BLVRA in' uterine part of Lushi chickens and Gushi chickens, thus discover the formation mechanism of green eggs. To conduct the experiment, Lushi chickens laying green eggs and Gushi chickens laying brown eggs, all of which were 24- week aged, were randomly selected and the uterus of each chicken was collected. Then the total RNA was extracted and later reversely transcribed into cDNA. The pigment-associated genes, HO-1 and BLVRA were chosen to be the target genes, and GAPDH the reference genes. Finally, the transcription levels of HO- 1 and BLVRA in different breeds of chickens were analyzed using real-time fluorescent quantitation PCR. The results showed that the HO-1 mRNA expression level in the uterus of Lushi chickens was higher than that of Gushi chickens (P〈0.05), and that the BLVRA mRNA expression level in the uterus of Gushi chickens was significantly higher than that of Lushi chickens (P〈0.01).
