5,7-双羟色胺侧脑室注射对大鼠内侧前额叶皮层神经元兴奋性的影响

摘  要:本研究采用在体细胞外记录的方法,探讨了双侧侧脑室注射5,7-双羟色胺(5,7-dihydroxytryptamine,5,7-DHT)损毁大鼠脑内5-羟色胺(5-hydroxytryptamine,5-HT)神经元后,内侧前额叶皮层(medial prefrontal cortex,m PFC)锥体和中间神经元兴奋性的变化。实验结果显示:5,7-DHT注射到双侧侧脑室后第2周,m PFC和中缝背核内的5-HT水平较正常组显著下降。m PFC的锥体神经元的放电频率与正常组相比明显升高,爆发式放电显著增多,而中间神经元的平均放电频率显著降低,放电形式趋向不规则活动。结果提示5-HT能递质系统对m PFC神经元活动具有重要的调节作用。The present study is aimed to investigated the firing activity of pyramidal neurons and interneurons in the medial prefrontal cortex(m PFC) in rats with bilateral intraventricular injection of 5,7-dihydroxytryptamine(5,7-DHT) by using in vivo extracellular recording. The results showed that the injection of 5,7-DHT reduced the 5-hydroxytryptamine(5-HT) levels in the m PFC and dorsal raphe nucleus in the rats. The firing rate of m PFC pyramidal neurons in rats with 5,7-DHT injection was significantly higher than that of normal rats, and the firing pattern of these neurons also changed significantly towards a more burst-firing, while the injection decreased the firing rate of m PFC interneurons and changed the firing pattern of the interneurons towards a more irregular. These results indicate that the lesions of the serotonergic neurons lead to the changes in the firing activity of m PFC pyramidal neurons and interneurons, suggesting that serotonergic system plays an important role in the regulation of the neuronal activity in the m PFC.
