摘  要:目的观察白藜芦醇(resveratrol,Res)对重症急性胰腺炎(SAP)肺损伤大鼠树突状细胞(DC)的影响。方法 36只SD大鼠随机分为三组:假手术组、SAP模型组、Res治疗组(Res 10 mg/kg),每组各12只。各组SD大鼠于制模后12 h处死,行胰腺及肺组织病理学检查和评分。提取肺树突状细胞,流式细胞仪检测DC表型CD80表达,并检测IL-12的分泌水平。结果 Res组肺和胰腺组织病理学评分分别为(3.49±1.02)和(6.43±1.40),而SAP组分别为(6.12±2.02)和(11.00±2.58),Res组评分均显著降低(P<0.05)。SAP组DC细胞表面分子CD80阳性表达率为(45.3±6.7)%,而Res组为(20.1±3.2)%,显著低于SAP组,差异有统计学意义(P<0.05);Res组和SAP组DC分泌IL-12浓度分别为(1 676.5±121.8)g/L和(4 436.4±169.8)g/L,Res组IL-12浓度显著低于SAP组,差异有统计学意义(P<0.05)。结论 Res能显著减轻SAP时肺和胰腺组织损伤,其机制与抑制肺DC活化有关。Objective To investigate the effect of resveratrol (Res) on dendritic cell in lung of rats with severe acute pancreatitis associated lung injury.Methods Thirty-six rats were divided into three groups:sham group,SAP model group and Res treated group.Rats were sacrificed at 12 h after inducing SAP model.The pathological changes of pancreas and lung were observed,and the level of IL-12 was examined by ELISA method.Results Histopathology scores of lung and pancreas in Res group were significantly lower than that in SAP group [(3.49 ± 1.02) and (6.43 ± 1.4),(6.12 ± 2.02) and (11.00 t 2.58),P<0.05)].The positive rate of the phenotype of CD80 in Res group was (20.1 ± 3.2) %,which was significantly lower than that in SAP group [(45.3 ± 6.7)%,P <0.05].The level of the IL-12 in Res group was (1 676.5 ± 121.8) g/L,which was significantly lower than that in SAP group [(4 436.4 ± 169.8) g/L,P<0.05].Conclusion Resveratrol can relieve the injury of pancreas and lung in SAP,the mechanism of which may be related with the inhibitory effects of Res on activation of dendritic cell.
