摘  要:目的 利用1H核磁共振代谢组学技术探讨干舱饱和暴露对人体代谢模式的影响.方法 7名健康男性潜水员参加干舱250 m氦氧饱和潜水实验,在实验前、中、后不同阶段取晨尿,应用600MHz核磁共振谱仪采集尿样谱图,应用SIMCA-P+12.0软件进行模式识别分析.结果 250 m压力氦氧逗留、减压及暴露全程对参试人员的代谢模式产生明显影响,筛选出尿液差异代谢物13个,分别是肌酸、肌酐、葡萄糖、柠檬酸、α-酮戊二酸、乳酸、乙酰乙酸、乙酸、苯丙氨酸、异亮氨酸、N-氧化三甲胺、二甲胺、马尿酸,主要反映机体的能量代谢、氨基酸代谢、胺类代谢和肾脏功能.结论 核磁共振谱仪代谢指纹提供了250 m干舱氦氧饱和暴露引起的内源性差异代谢物信息.Objective To investigate the effects of 250 m helium-oxygen (heliox) saturation dive on the mode of human metabolism by high-resolution nuclear magnetic resonance spectroscopy.Methods Seven male divers performed the 250 m heliox saturation dive by using the dry diving simulator.Urine morning samples were collected at various stages,before dive,during dive and after dive.Urine spectra were acquired by using the 600 MHz nuclear magnetic resonance (NMR),and then mode identification was performed by SIMCA-P + 12.0 software.Results Stay at the storage depth,decompression and the whole process of saturation exposure at a pressure of 250 m exerted significant effects on the metabolismof divers.From urine samples,13 metabolites were screened (P < 0.05).They were creatine,creatitine,glucose,citric acid,aketoglutarate,lactate,acetoacetate,acetate,phenylalanine,isoleucine,N-oxide trimethylamine,dimethylamine and hippuric acid,which could reflect energy metabolism,amino acid metabolism,amine metabolism and kidney function.Conclusions NMR-based metabonomic analysis could provide information on endogenous metabolites which characterized changes in energy supply form induced by the 250 m heliox saturation exposure in the dry chamber.
