金银花与山银花对致敏大鼠外周血单个核细胞的影响

摘　　要：目的免疫豚鼠获取抗血清,观察金银花与山银花中间体等对豚鼠的致敏性及其对大鼠外周血单个核细胞(PBMC)增殖的影响。方法金银花和山银花各中间体等免疫豚鼠并观察豚鼠致敏情况,收集致敏后豚鼠血清,将其与大鼠PBMC细胞共孵育24 h,建立致敏细胞模型。金银花与山银花各中间体等与致敏细胞共培养24 h,CCK-8法检测大鼠PBMC的增殖情况。结果金银花中间体1、2发生豚鼠过敏反应;山银花中间体1、2发生豚鼠类过敏和过敏反应;金银花与山银花中间体等均抑制体外培养的大鼠PBMC的增殖,但两者相比金银花抑制率偏低。结论参照热毒宁注射液单味模拟药物的制备工艺,金银花对豚鼠的致敏性、对大鼠PBMC的抑制率低于山银花。Objective To observe the sensitization of the guinea pig and multiplication of the rat PBMC after getting immunized guinea pig antiserum with the intermediates of Lonicera japonica and Lonicera confus. Methods After observing the allergic reaction of the guinea pigs which were sensitized with the intermediates of Lonicera japonica and Lonicera confusa,the blood serum was collected,then the PBMC of rat was cultured with the blood serum for 24 hours and the cell sensitization model was built. The multiplication of the rat PBMC was detected by CCK-8 technique after the intermediates of Lonicera japonica and Lonicera confusa were cultured with the sensitized cell for 24 hours. Results The one and two intermediates of Lonicera japonica were discovered allergy; The one and two intermediates of Lonicera confusa occurred anaphylactoid reaction and allergy in guinea pig; The intermediates of Lonicera japonica and Lonicera confusa both inhibited the proliferation of the rat PBMC. However,the inhibition rate of the intermediates of Lonicera japonica was lower compared with that of Lonicera confusa. Conclusion The sensitization of Lonicera japonica in guinea pigs and inhibition ratio of rat PBMC were lower than those of Lonicera confuse with reference to the drug preparation technology of single simulation of Reduning Injection.
