摘  要:目的研究顺铂致大鼠急性肾损伤(AKI)模型中AKI标志物中性粒细胞明胶酶相关脂质运载蛋白(NGAL)变化及还原型谷胱甘肽干预疗效。方法 66只健康雄性Wistar大鼠为研究对象,随机数字法随机分为盐水对照组(CN组),顺铂模型组(CP组)及还原型谷胱甘肽干预组(GI组),CP组为腹腔单次注射顺铂10mg/kg,据给药后各时间点不同分为5组;GI组为应用顺铂前腹腔注射还原型谷胱甘肽800 mg/kg,同样据给药时间不同分为5组;CN组为腹腔注射同顺铂组等容量生理盐水。观察用药后12、24、48、96 h及144 h各组大鼠血清肌酐(Scr)及用ELISA法检测血、尿NGAL变化情况。结果同CN组相比,CP组12 h血、尿NGAL开始升高,48 h达峰值,Scr 48 h开始升高,144 h达峰值。GI组同CP组比较,减轻了血、尿NGAL的升高。结论 CDDP致大鼠AKI早期存在血、尿NGAL的升高,还原型谷胱甘肽可一定程度上减轻NGAL的升高,可能为防治顺铂致急性肾损伤的有效药物。[ Objective ] To observe the effect of reduced glutathione on serum and urinary concentration of neutrophil gelatinase-associated lipocalin (NGAL) in the rodent model of cisplatin induced acute kidney injury (AKI). [ Methods ] 66 male wistar rats were randomly assigned to three groups. The rats of CP group were injected once intraperitoneal with cisplatin at 10 mg/kg, the rats of CN group were injected intraperitoneal with same volume of normal saline vehicle, the GI group were injected intraperitoneal with reduced glutathione at 800 mg/kg before cis- platin injection. Rats were euthanized at 12, 24, 48, 96 and 144 hours (n =6 in the CP group at each time point and n =6 in the GI group at each time point). The blood and urinary samples were analyzed at each time point. [Results ] Significant elevations of NGAL in serum and urine were noted by 12 hours in the CP group versus CN group, peak levels were seen on 48 hours(P 〈0.05). A significant elevation of serum creatinine was noted by 48 hours in the CP group versus CN group, peak level was seen on 144 hours(P 〈0.05). Compared with CP group, the concentrations of serum and urinary NGAL in GI 48 hours group decreased significantly. [ Conclusions ] The levels of NGAL in serum and urine were increased significantly at early phase of eisplatin induced AKI, reduced glutathione could at- tenuate the increases of NGAL in serum and urine, which may be an effective way against cisplatin induced AKI.
