摘  要:目的探讨碱性成纤维生长因子(b FGF)在脂肪干细胞(ADSC)治疗心肌梗死中的作用。方法从65只SD大鼠中提取、分离、培养、鉴定ADSC后,用细胞分化诱导液培养评价ADSC分化能力。将成功建立心肌梗死模型的60只大鼠随机分4组:磷酸盐缓冲液(PBS)组(n=15)、b FGF组(n=15)、ADSC组(n=15)及ADSC+b FGF组(n=15)。建立心肌梗死1周后分别注射PBS、b FGF、ADSC及ADSC+b FGF。移植4周后用心脏超声测量心肌梗死大鼠的LVEF,取大鼠心脏检测左心室梗死面积、微血管数量及ADSC分化情况。结果成功分离ADSC,流式细胞分析显示大多数ADSC表达CD29和CD90,不表达CD34和CD45。ADSC能被诱导分化成成脂、成骨细胞。ADSC+b FGF组及ADSC组的LVEF高于PBS组和b FGF组(P<0.01),ADSC+b FGF组的LVEF高于ADSC组(P<0.01),PBS组和b FGF组的LVEF比较差异无统计学意义(P=0.33)。ADSC+b FGF组及ADSC组的梗死面积小于PBS组和b FGF组(P<0.01),ADSC+b FGF组的梗死面积小于ADSC组(P<0.01),PBS组和b FGF组的梗死面积比较差异无统计学意义(P=0.28)。ADSC+b FGF组的微血管数量最多(P<0.01)。ADSC可分化为c Tn T阳性细胞、SMA阳性细胞及v WAg阳性细胞。结论 b FGF可促进ADSC分化成心肌细胞和新生微血管,改善心脏功能。Objective To study the effects of basic fibroblast growth factor (bFGF)in the treatment of myocardial infarction with adipose derived stem cells (ADSCs).Methods ADSCs were extracted,isolated,cultured and verified from 65 Sprague Dawley rats.The differentiation potential of ADSCs into adipogenic cells was evaluated.Sixty myocardial infarction (MI)rat models were successfully established and randomly assigned into four groups:PBS group (n =15),bFGF group (n =15),ADSCs group (n =15)and ADSCs +bFGF group (n =15).PBS,bFGF,ADSCs and ADSCs +bFGF were injected into the left ventricular wall of MI rats at one week after establishment.Left ventricular ejection fraction (LVEF),MI area,microvessel densi-ty and ADSC differentiation were evaluated at four weeks following transplantation.Results ADSCs were suc-cessfully isolated and most ADSCs showed positive CD90 and CD29,whereas negative CD45 and CD34 by fluo-rescence-activated cell sorting (FACS).ADSCs could be induced to differentiate into adipogenic and osteogen-ic cells.LVEF in the ADSC +bFGF and ADSC groups was significantly higher compared with those in the PBS and bFGF groups (all P 〈0.01).LVEF in the ADSC +bFGF group was significantly higher than that in the ADSC group (P 〈0.01).LVEF did not significantly differ between the PBS and bFGF groups (P =0.33). MI area in the ADSC +bFGF and ADSC groups was significantly smaller than those in the PBS and bFGF groups (all P 〈0.01).MI area did not significantly differ between the PBS and bFGF groups (P =0.28). Microvessel density in the ADSC +bFGF group was largest among different groups (P 〈0.01).ADSCs could&amp;nbsp;differentiate into cTnT positive,SMA positive and vWAg positive cells.Conclusion bFGF was able to im-prove heart function by promoting ADSCs to differentiate into myocardial cells and microvessels.
