摘 要:目的:建立非水毛细管电泳法(non-aqueous capillary electrophoresis,NACE)快速测定萝芙木中育亨宾的含量。方法:运用非水毛细管电泳方法,以小檗碱作为内标,缓冲液为40 mmol·L-1乙酸铵-2.0%醋酸-20%乙腈的甲醇液,检测波长220 nm,分离电压25 k V,柱温20℃,压力进样:50 mbar×3 s。结果:方法最低检测浓度为1.0μg·m L^-1,线性范围10-100μg·m L^-1,r=0.996,线性关系良好。平均回收率为97.8%,RSD为1.74%。结论:本法用于萝芙木中育亨宾的含量测定快速、准确且灵敏。Objective To develop a rapid method for the determination of yohimbine in Rauvolfia verticillata by nonaqueous capillary electrophoresis (NCAE). Methods A NACE method has been employed using berberine as an internal standard. The separation conditions were 40 mmol · L^-1 ammonium acetate (methanol medium) ,2.0% glacial acetic acid and 20% acetonitrile in methanol as running buffer, UV detection was set at 220 nm,separation voltage was 25 kV,column temperature was 20 ℃ and pressure injection was 50 mbar × 3 s. Results The limits of detection was 1.0 · mL^-1. The concent ration of yohimbine showed a linear plot in the range of 10 - 100 μg · mL^-1 ( r =0.996). The main recovery was found to be 97. 8%. The variation was 1.74%. Conclusion This method is rapid and accurate for the quantitative analysis of yohim- bine in Rauvolfia verticillata.
