人脑胶质瘤MMP2/TIMP2与肿瘤血管的相关性分析

摘　　要：目的探讨人脑胶质瘤中的基质金属蛋白酶2(MMP2)、金属蛋白酶2组织抑制剂(TIMP2)与血管内皮生长因子(VEGF)及其受体(VEGFR)表达的相关性,分析其在肿瘤血管形成中的作用。方法应用免疫组化SP法检测40例脑胶质瘤及12例正常脑组织中MMP2、TIMP2、VEGF、VEGFR1、和VEGFR 2蛋白的表达。结果 MMP2、VEGF、VEGFR1和VEGFR2蛋白表达的免疫反应评分(IRS)在胶质瘤中明显升高,TIMP2蛋白表达的免疫反应评分(IRS)在胶质瘤中明显降低,差异有统计学意义(P<0.05);胶质瘤组中MMP2与VEGFR2、TIMP2蛋白表达的免疫反应分数显著相关(r=0.396,P<0.01;r=0.317,P<0.05)。结论 MMP2/TIMP2可能通过影响VEGFR-2介导的信号转导途径调控胶质瘤肿瘤血管生成。Objective To discuss the relationship between the expression of matrix metalloproteinase 2 （ MMP2 ） , tissue inhibitor of metalloproteinase-2 （ TIMP2 ） with vascular endothelial growth factor（VEGF） and vascular endothelial growth factor receptor（VEGFR）in human glioma.Methods The protein expression of MMP2, TIMP2, VEGF, VEGFR1 and VEGFR2 was detected through immunohistochemistry in 40 patients with glioma and 12 with normal brain tissue. Results Immunoreactivity score of MMP2,VEGF,VEGFR1 and VEGFR2 was increased in glioma with the decrease of TIMP2（P〈0.05）.Positive correlation between MMP2 and VEGFR2,TIMP2（r=0.396,P〈0.01;r=0.317,P〈0.05）showed significant level.Conclusion MMP2/TIMP2 may be implicated in tumor angiogenesis through the VEGF-VEGFR2 pathway.
