摘  要:目的观察骨痹颗粒对骨关节炎大鼠的保护作用,初步探讨其作用机制。方法 SD大鼠随机分为正常对照组、模型组、假手术组、塞来昔布组及骨痹颗粒高、中、低3个剂量组;采用改良Hulth法建立大鼠骨关节炎模型,连续灌胃给药8周后,观察骨痹颗粒对大鼠血清中炎症因子、基质金属蛋白酶及抑制剂及病理组织学变化的影响。结果骨痹颗粒可显著抑制炎症因子IL-1及TNF-α的释放(P〈0.05-0.01),降低MMP-1及MMP-3水平(P〈0.05-0.01),提高TIMP-1水平(P〈0.05),改善软骨病理变化,降低Mankin's评分(P〈0.05)。结论骨痹颗粒对大鼠骨关节炎具有良好的防治作用,其机制与抑制炎症因子释放、调整基质合成与代谢的平衡有关。OBJECTIVE To investigate the protective effect of Gubi granules on osteoarthritis in rats and its mechanism. METH- ODS SD rats were randomly divided into normal control group, model group, sham-operation group, celecoxib group, as well as high, medium and low dose groups of Gubi granules. Osteoarthritis models of rats were established by improved Hulth's modeling method. After 8 weeks of continuous intragastric administration, the effects of Guhi granules on inflammatory serum cytokines in rats, matrix metalloproteinase and histopathological changes were observed. RESULIS Gubi granules significantly inhibited the release of inflammatory cytokines IL-1β and TNF-a(P〈0.05-0.01), reduced the level of MMP-1 and MMP-3 (P〈0.05-0.01), increased the level of TIMP-1 (P〈0.05), improved pathological changes of osteoarthritis and reduced Mankin's score(P〈0.05). CONCLUSION Gubi granules have excellent preventive and therapeutic effect on osteoarthritis, and its mechanism is related to the inhibition of in- flammatory cytokine release and the adjustment of matrix synthesis and metabolism balance.
