摘  要:目的:优选葛根中葛根素的闪式提取工艺。方法:在单因素试验基础上,以葛根素提取率为评价指标,采用正交试验法考察乙醇体积分数、料液比、提取电压、提取时间对葛根中葛根素提取工艺的影响。采用HPLC测定葛根素含量,流动相甲醇-水(25∶75),检测波长250 nm。结果:最佳提取工艺为加15倍量50%乙醇于90 V提取2 min;葛根素提取率37.42 mg·g-1,转移率88.89%。结论:优选的工艺稳定可行、重复性好、提取时间短,可作为葛根中葛根素的工业化生产工艺参数。Objective:To optimize flash extraction process of puerarin from Puerariae Lobatae Radix.Method:Based on single tests,taking extracting rate of puerarin as index,orthogonal test was adopted to optimize flash extraction process with ethanol concentration,solid-liquid ratio,extraction voltage and time as factors.HPLC was employed to determine the content of puerarin with mobile phase of methanol-water (25 ∶ 75) and detection wavelength at 250 nm.Result:Optimum extraction technology was as following:extracted 2 min with 15 times the amount of 50% ethanol at 90 V.Under these conditions,extracting rate of puerarin was 37.42 mg ·g^-1 and its transfer rate was 88.89%.Conclusion:This optimized process is stable and reproducible with short extracting time,it can be used as process parameters for industrial production of puerarin from Puerariae Lobatae Radix.
