不同功效中药对裸鼠肺癌皮下移植瘤及转移相关微小RNA的影响  

Influence of Chinese Medicines with Different Effects on Subcutaneous Tumors Generated from Lung Cancer Cells and Metastasis Associated MicroRNAs in Nude Mice

摘　　要：目的观察益气养阴、健脾温肾、软坚散结中药对裸鼠人肺癌NCI-H460细胞皮下移植瘤的影响及其可能作用机制。方法将人非小细胞肺癌细胞株NCI-H460接种于32只裸鼠皮下,成瘤后随机分为健脾温肾组、益气养阴组、软坚散结组和生理盐水组,每组8只。接种1个月起各组小鼠分别灌胃给予相应药物,健脾温肾方、益气养阴方、软坚散结方药物浓度分别为1.1、1.9、1.8 g/ml,每日1次,每次0.3ml,共15天。检测各组荷瘤小鼠肿瘤体积和重量,计算抑瘤率,PCR检测肺癌皮下移植瘤中MicroRNA-21、MicroRNA-218的表达。结果给药10天时各组小鼠瘤体积均较给药前明显增大,给药15天时益气养阴组、软坚散结组、生理盐水组小鼠瘤体积较给药前增大(P<0.05)。温肾健脾组、益气养阴组、软坚散结组肿瘤抑制率分别为9.9%、16.3%、11.8%,各组抑制率比较差异无统计学意义(P>0.05)。益气养阴组瘤重、MicroRNA-21的表达量均明显低于生理盐水组(P<0.05)。结论益气养阴方可以抑制裸鼠皮下移植瘤的生长,下调致癌基因相关微小RNA的表达可能是其作用机制之一。Objective To observe the impact of Chinese medicines with function to tonify qi and nourish yin（ YQYY）,strengthen spleen and warm kidney（ JPWS）,and soften hardness and disperse nodules（ RJSJ） on subcutaneous tumor transplanted from human lung cancer NCI-H460 cells in nude mice,and to discuss its possible mechanism. Methods The human non-small cell lung cancer（ NSCLC） cell line H460 was inoculated subcutaneously into 32 nude mice. After tumor formation,all mice were randomly divided into a JPWS group,a YQYY group,a RJSJ group,and a saline group,with 8 in each. The concentration of JPWS,YQYY,and RJSJ were 1. 1 g / ml,1. 9g / ml,and 1. 8 g / ml,respectively. One month after inoculation all mice were given corresponding medicinals by gastric administration,0. 3 ml each time,once a day,for 15 days. Tumor volume and weight in tumor-bearing mice were measured,inhibition rate was calculated,and expression of MicroRNA21 and MicroRNA218 was detected by PCR in each group. Results On Day 10 after administration,the tumor volume of mice in all group increased significantly compared with before medication,and on Day 15 the tumor volume of mice in the YQYY group,RJSJ group,and saline group increased compared with before treatment（ P〈0. 05）. The tumor inhibition rates in the JPWS group,YQYY group,and RJSJ group were 9. 9%,16. 3%,and 11. 8%,respectively,with no significant differences among groups（ P 〉0. 05）. The tumor weight and MicroRNA-21 expression levels in the YQYY group were significantly lower than that in the saline group（ P〈0. 05）. Conclusion Medicines with the effect of tonifying qi and nourishing yin can inhibit tumor growth in nude mice. Down-regulating the expression of oncogenes related MicroRNA may be one of the possible mechanisms.
