摘 要:目的:建立橘核的高效液相色谱(HPLC)指纹图谱。方法:采用HPLC法。色谱柱为Dikma C18,流动相为乙腈-0.3%磷酸水溶液(梯度洗脱),柱温为30℃,流速为1.0 ml/min,检测波长为210 nm,进样量为20μl。采用《中药色谱指纹图谱相似度评价系统》对18个批次的橘核进行分析,计算18批样品共有指纹峰的相对保留时间及相对峰面积,并采用类平均法为聚类方法对样品进行归类。结果:橘核药材的指纹图谱共包含有11个共有峰,其中9号峰为柠檬苦素。18个批次橘核药材与对照图谱的相似度均>0.90。聚类分析结果显示,市售橘核样品和自采橘核样品归为两类,表明采集方式对橘核药材各成分的含量有影响。结论:该研究方法和所获得的指纹图谱具有较好的重复性,可作为橘核药材的鉴定和质量评价依据。OBJECTIVE: To establish HPLC fingerprint of active ingredients in Citrus reticulate. METHODS: HPLC method was adopted. The determination was performed on Dikma C18 column with mobile phase consisted of acetonitrile-0.3% phosphoric acid (gradient elution) at the flow rate of 1.0 ml/min. The detection wavelength was 210 nm and column temperature was set at 30 ℃, The sample size was 20 μl. TCM chromatogram fingerprint similarity evaluation system was used to analyze 18 batches of C. reticulate, and calculate relative retention time and relative peak area of common fingerprint peak of 18 batches of sample. The samples were classified by group-average method of clustering analysis. RESULTS: There were 11 common peaks of fingerprint, among which the 9th peak was limonin. The similarity of 18 batches of C. reticulate to control chromatogram was above 0.90. Results of cluster analysis divided C. reticulate into commercially available sample and self-served sample, which indicated collection means affected the content of each component in C. reticulate. CONCLUSIONS: The method and fingerprint show good reproducibility, and can provide reference for identification and quality evaluation of C. reticulate.
