摘  要:目的:考察岗梅中皂苷的提取方法并建立不同部位中总皂苷的含量测定方法。方法:采用分光光度法测定和比较不同提取方法及不同部位中得到的总皂苷含量。以18-去氢乌苏酸为对照品,香草醛、冰醋酸和高氯酸为显色剂,在555 nm波长处测定其吸光度。结果:采用50%乙醇、回流1.5 h的方法提取较完全。18-去氢乌苏酸在20.3~121.8 ug/m L质量浓度范围内线性关系良好,回归方程Y=0.0083X-0.02,r=0.9996,平均加样回收率为98.28%,RSD%为1.92%(n=6),岗梅根、茎、叶中总皂苷的含量分别为3.93%、3.31%、3.38%。结论:分光光度法简便,准确,重现性好,可用于岗梅总皂苷的含量测定。岗梅根、茎、叶中均含有一定量的皂苷。Objective: To study the extraction method and establish the method for the content determination in different parts of total saponins in Gangmei (Ilex asprella) .Methods: Ultraviolet spectrophotometry was applied to compare the content of total saponins extracted by different extraction methods and in different parts. Vanillic aldehyde,glacial acetic acid and perchloric acid were used as color-display reagent;18-dehydro urane acid as reference substance. The detection wavelength was set at 555 nm. Results: Using 50% alcohol and refluxed for one and half an hour can entirely extract. The linear range of 18-dehydro u- rane acid was 20.3-121.8μg/mL(r=0.9996), Y=0.0083X-0.02,r=0.9996 with an average recovery of 98.28%, RSD=-1.92% (n=6). The total saponins content in root; stem and leaf was 3.93%, 3.31% and 3.38% respectively. Conclusion: The method was accu- racy, simple and rapid. The reproducibility was good, so the method was suitable for the content determination of total saponins in Gangmei (Ilex asprella). There is certain content of saponins in root, stem and leaf of Gangmei (Ilex asprella).
