摘  要:采集关中某羊场奶山羊的血液样品,以提取的血液基因组DNA为模板对弓形虫ITS1及529bp基因进行PCR扩增、测序,并对测序结果进行比对,基于529bp重复序列构建系统进化树。序列分析结果表明,待测样品与GenBank中弓形虫RH虫株的ITS1序列(AY259044)完全一致,与529bp重复序列(FJ656209.1)存在差异;基于529bp重复序列构建的系统进化树分析结果显示,待测样品弓形虫与RH虫株遗传距离最近。该结果为奶山羊弓形虫病的分子流行病学、诊断方法的建立等方面的研究奠定了基础。Blood samples of dairy goats was collected from a farm in Guanzhong area so that the genomic DNA was extracted as the template for PCR amplification, sequencing of Toxoplasma gondii ITS1 and 529 bp repetitive sequence, and analyzing the results by BLAST, then the evolutionary tree was constructed based on 529 bp repeat sequence. The results of sequence analysis showed that the test samples were identical to RH strain ITS1 sequence (AY259044) of T. gondii, and different from 529bp repetitive sequence (FJ656209.1), the evolutionary tree of 529 bp repetitive sequence suggested that the genetic distance between test samples and T. gondii RH strain is the nearest. The results lay the foundations for the study of molecular epidemiology and diagnosis method establishment of toxoplasmosis in dairy goat.
