摘 要:目的研究孔石莼(Ulva pertusa Kjellm)凝集素的最佳提取条件。方法通过研究各因素(缓冲液、料液比、浸泡时间、烘干温度)对孔石莼(Ulva pertusa Kjellm)凝集素(lectin)(UPL)提取液血凝活性的影响,得出每个因素的最佳提取效果。从各个单因素中选出提取的最适条件及与最适条件相临的2个水平(共3个水平)进行4因素3水平的正交试验。结果温度为50℃、缓冲液为pH 7.2PBS、料液比为1∶15、浸泡时间为15h。结论相比较其它条件,这一组合对孔石莼提取效果最好。Objective To provide best theory basis for extraction process of lectin from Ulva pertusa Kjellm. Methods This paper aimed to explore a kind of optimal combination on hemagglutinating activity of Ulva pertusa Kjellm lectin. First of all, the influence of the different factors including buffer (contain different pH), material ratio, soaking time, drying temperature were evaluated to study the best effect on extraction of Ulva pertusa Kjellm lectin of each factors. Then the optimal conditions of extraction and two abutting conditions were selected to conduct orthogonal test. Every single factor from selected the optimal conditions and optimum condition that faced two levels (of three levels) were conducted the orthogonal test. Results The best optimal combination was that the temperature was 50 % and the buffer pH was 7.2 and the material Conclusion Compared with other conditions, this Kjellm lectin. -water ratio was 1 - 15 and the soaking time was 15 h. combination was very good to extract Ulva pertusa
