摘  要:目的观察脂氧素A4(LXA4)对小鼠重症急性胰腺炎相关肺损伤的作用。方法将30只ICR小鼠随机分为对照组即假手术组、SAP组及LXA4治疗组,每组10只小鼠。SAP组及LXA4组小鼠采用逆行胰胆管注射法建立重症急性胰腺炎模型,术后1 h,LXA4小鼠尾静脉给予LXA4 0.1 mg/kg,建模后24 h处死小鼠,检测肺湿干重比,ELISA法检测血清TNF-α、IL-10,观察小鼠肺组织病理变化并评分,比色法检测肺组织髓过氧化物酶(MPO),TBA法检测肺组织丙二醛(MDA)含量及Western Blot检测肺组织核转录因子-κB p65、IκBα的表达情况。结果与SAP组相比,LXA4治疗组血清TNF-α,肺组织病理评分、肺湿干重比、MPO活性、MDA含量及NF-κB p65表达均有所降低,血清IL-10及肺组织IκBα表达上升,差异有统计学意义(P<0.05)。结论 LXA4对小鼠重症胰腺炎相关肺损伤有保护作用,其机制可能是通过下调NF-κB的表达实现的。Objective To observe the protective effect of lipoxin A4( LXA4) on acute lung injury associated with acute pancreatitis in mice. Methods 30 ICR mouse were randomly divided into control group,SAP group and LXA4 group,with 10 mouse in each group. The retrograde pancreatic duct injection was performed in the SAP group and LXA4 group to establish the model of severe acute pancreatitis,1 hour after the operation; LXA4 0. 1 mg / kg was administrated via tail vein,all the mouse was sacrificed within 24 hours after modeling. The wet / dry lung weight ratio was detected; The levels of serum TNF-α and IL-10 were tested by ELISA; pathological changes of lung tissues were observed and scored,myeloperoxidase( MPO) activity of lung tissue was detected by colorimetry,and the content of malondialdehyde( MDA) in lung tissue was measured by TBA,the expression of nuclear factor-κBp65 and IκBα in lung tissue was measured by Western Blot. Results As compared with SAP group,,the level of serum TNF-α,histopathological scoring of lung tissue,wet / dry lung weight ratio,MPO activity,MDA content and the expression of NF-κBp65 in the LXA4 treatment group was decreased,IL-10 of lung tissue and serum IκBα rose,and the difference was statistically significant( P〈0. 05).Conclusion LXA4 shows a protective effect on the acute lung injury associated with acute pancreatitis in mice,the mechanism may be realized through down-regulation of NF-κB.
