通心络胶囊对糖尿病小鼠视网膜组织LN和FN表达的影响

摘　　要：目的探讨通心络胶囊对糖尿病小鼠视网膜组织纤维粘连蛋白(FN)、层粘连蛋白(LN)表达的影响。方法 40只KK/Upj-Ay小鼠分为模型组与通心络高、中、低剂量(4、2、1 g/kg)组,每组10只,灌胃途径给药,持续12周;另取10只C57BL/6小鼠作正常对照组。应用RT-PCR法检测各组小鼠视网膜组织中FN、LN的mRNA表达;Western blot法检测视网膜组织中FN、LN蛋白的表达。结果与对照组相比,模型组小鼠视网膜组织LN、FN mRNA和蛋白表达均明显增高(P<0.05),通心络低、中、高各剂量组LN、FN mRNA和蛋白表达水平均较模型组明显下降(P<0.05)。结论通心络胶囊可通过抑制糖尿病小鼠视网膜组织LN、FN的表达从而改善视网膜病变。Objective To explore the effect of Tongxinluo capsule on the expression of fibronectin （ FN） and laminin （LN） in retina of KK/Upj-Ay mice.Methods Forty KK/Upj-Ay mice were randomly divided into 4 groups：model group, Tongxinluo low-dose group （1g/kg）,median-dose group （2g/kg）,high-dose （4g/kg）group.The mice were given medicines intragastrically once a day for consecutive 12 weeks.The other 10 C57BL/6 mice were enrolled as control group .The expression levels of FN and LN mRNA in retina were detected by Real time PCR ,and the contents of protein were determined by Western blotting .Results As compared with those in control group ,the expression levels of LN ,FN mRNA and protein in retina in model group were significantly increased （ P 〈0.05）,however,which in Tongxinluo low-dose group,median-dose group,high-dose group were significantly decreased ,as compared with those in model group （ P 〈0.05）.Conclusion Tongxinluo capsule may improve diabetic retinopathy by inhibiting the expression of LN and FN in retina tissue of diabetic mice.
