脉君安片对自发性高血压大鼠血压和电解质的影响

摘　　要：目的研究脉君安片及其组方对自发性高血压大鼠（SHR）血压和电解质的影响。方法 ① 24只SHR随机分为模型组（10.0 ml/kg的0.5%羧甲基纤维素钠）、脉君安组（脉君安片2.1 g/kg）、钩藤＋葛根组（钩藤0.8 g/kg和葛根1.8 g/kg）、氢氯噻嗪组（氢氯噻嗪10.0 mg/kg）,6只Wistar-Kyoto（WKY）大鼠作为对照组（10.0 ml/kg的0.5%羧甲基纤维素钠）。每天均予固定时间给药,每周固定1天（周一）给药1 h后使用BP-98A动物无创血压计测量血压,连续给药13周。腹主动脉取血清检测电解质（Na^＋、K^＋、Cl^-）水平;② 24只SD大鼠随机分为脉君安组（脉君安片4.2 g/kg）、钩藤＋葛根组（钩藤1.6 g/kg和葛根3.6 g/kg）、氢氯噻嗪组（氢氯噻嗪20.0 mg/kg）和对照组（灌胃等量生理盐水）,测量给药后0~2 h和2~4 h的尿液量。结果脉君安给药第1周可引起收缩压（SBP）下降24.4 mmHg,舒张压（DBP）下降19.3 mmHg,给药13周,SHR的SBP维持在158.9~169.4 mmHg,DBP维持在112.8~136.3 mmHg,氢氯噻嗪组和钩藤＋葛根组SHR的血压均低于模型组;在0~2 h内脉君安和氢氯噻嗪均可显著增加水负荷大鼠的排尿量,而钩藤＋葛根组不仅没有利尿作用,却显著减少2~4 h的尿量;但与氢氯噻嗪组可明显降低大鼠血钾水平相比,脉君安组和钩藤＋葛根组未见降低大鼠血钾的作用。结论脉君安片及其各组方均有一定的降压作用,其中钩藤和葛根成份对氢氯噻嗪的利尿作用并无影响,并可改善氢氯噻嗪引起的钾离子丢失。Objective To investigate the effects of Maijun＇an tablets on blood pressure and serum electrolytes in spontaneous hypertensive rats（SHRs）. Methods ①SHRs（n=24） and Wistar-Kyoto rats（n=6） were randomly divided into control group and drug administration group. SHRs in drug administration group were orally given with Maijun＇an tablets 2.1 g/kg, hydrochlorothiazide 10.0 mg/kg, puerarin 1.8 g/kg and rhynchophylla 0.8 g/kg, and Wistar-Kyoto rats in control group were orally dosed with 0.5% SHRs 10.0 ml/kg. BP-98 A noninvasive sphygmomanometer was used to measure blood pressure of SHR after 1 h with administration. Animals were treated with Maijun＇an or its components for 3 weeks. At the end of dosing day, animals were sacrificed for serum electrolytes analyses. ② SD rats（n=24） were randomly divided into control group and drug administration group. Rats in drug administration group were orally given with Maijun＇an tablets 4.2 g/kg, hydrochlorothiazide 20.0 mg/kg, puerarin 3.6 g/kg and rhynchophylla 1.6 g/kg, while rats in control group were orally dosed with equal amount of physiological saline. The urine during the interval of 0~2 h or 2~4 h was collected respectively, and the urine volume at the corresponding time interval was compared. Results Treatment with Maijun＇an tablets had a significant depressurization effect in the first week after a 3-week treatment with Maijun＇an. The SBP of SHR can be maintained at 158.9~169.4 mm Hg, DBP at 112.8~136.3 mm Hg. In the diuretic experiment, the treatment with Maijun＇an tablets or hydrochlorothiazide could increase the 2 h urinary amount. Compared with the hydrochlorothiazide group, the potassium levels in the Maijun＇an tablets group and puerarin＋rhynchophylla group were increased significantly. Conclusion The puerarin and rhynchophylla components in Maijun＇an tablets have no effects on the diuretic effects by hydrochlorothiazide,but they can improve the potassium loss induced by hydrochlorothiazide.
