摘  要:目的研究人肌腱成纤维细胞(tendon derived fibroblasts,TDFs)在聚乳酸(poly-L-lactic acid,PLLA)纳米纤维支架及聚乳酸/Ⅰ型胶原蛋白(PLLA/Col-Ⅰ)纳米纤维支架上的生长情况及相关基因的表达情况。方法将TDFs分别种植于空白盖玻片(空白组)、载有PLLA纳米纤维支架的盖玻片(PLLA组)和载有PLLA/Col-Ⅰ纳米纤维支架的盖玻片上(PLLA/Col-Ⅰ组),观察细胞生长情况,并检测Ⅰ、Ⅲ、Ⅹ型胶原蛋白(Col-Ⅰ、Col-Ⅲ、Col-Ⅹ)以及整合素相关基因黏着斑激酶(focal adhesion kinase,FAK)的基因表达情况。结果 PLLA组细胞生长速度低于空白组和PLLA/Col-Ⅰ组,而PLLA/Col-Ⅰ组和空白组相比,细胞生长速度差异无统计学意义。和PLLA组及空白组相比,PLLA/Col-Ⅰ组中Col-Ⅰ、Col-Ⅲ、Col-Ⅹ以及FAK基因表达水平显著升高,PLLA组和空白组无明显区别。结论 PLLA/Col-Ⅰ纳米纤维支架对TDFs的生长有促进作用,且有利于Col-Ⅰ、Col-Ⅲ、Col-Ⅹ及FAK基因的表达,能为肌腱修复提供一种理想的组织工程支架材料。Objective To explore the effects of PLLA nanofibers and PLLA/Col- Ⅰ nanofibers on human tendon derived fibroblasts (TDFs), and the related gene expression. Methods TDFs were respectively cultured on the cover slip (blank group), PLLA nanofibers (PLLA group) and PLLA/ Col- Ⅰ nanofibers (PLLA/Col- Ⅰ group). The growth of TDFs, and the gene expression of collagens Ⅰ , Ⅲ , Ⅹ and FAK during the course of culture were analyzed. Results The growth of TDFs in PLLA group was significantly inhibited as compared with blank group and PLLA/Col-Ⅰgroup. The gene expression of collagen Ⅰ , collagen Ⅲ and collagen Ⅹ , and FAK was remarkably up-regulated in PLLA/ Col- Ⅰ group as compared with PLLA group and blank group, but there was no significant difference between PLLA group and blank group. Conclusion This study indicates that PLLA/Col-Ⅰ nanofibers can promote the growth of TDFs, and can enhance the gene expression of collagen Ⅰ , collagen Ⅲ and collagen Ⅹ remarkably, as well as the gene expression of FAK. PLLA/Col- Ⅰ nanofibers may be an ideal scaffold of tissue engineering technology in the repair of tendon.
