摘 要:目的验证胶体金免疫层析法应用于环境水样本中霍乱检测的特异性、灵敏性和实用性,寻求简便、可靠的环境水中霍乱检测方法。方法环境水样置36.5℃经6 h增菌培养后,用胶体金免疫层析法检测,同时用传统分离培养法,培养后的菌落用血清玻片凝集方法作为鉴别霍乱弧菌的金标准,且用统计学方法对这两种方法检测的结果进行比较。结果 747份水样中胶体金试验阳性19份,分离培养法培养出16株霍乱弧菌,经χ2检验,差异无统计学意义(χ2=0.61,P>0.05),表明胶体金免疫层析法与传统分离培养法检测外环境水中霍乱的结果之间差异无统计学意义。胶体金免疫层析法检测外环境水中霍乱的灵敏度为100.00%,特异性为99.59%,假阳性率为0.41%,假阴性率为0.00%,与传统分离培养法的结果一致率为99.60%。结论胶体金免疫层析法检测外环境水中霍乱的敏感度好(100.00%),特异性高(99.59%),与传统分离培养法的结果一致性较高,对外环境水中霍乱检测具有可靠性、简便性、实用性,适宜在基层作为辅助检测方法推广应用。Objective To validate the specificity, sensitivity and practicability of detection of vibrio cholerae in outer environment water by colloidal gold immune chromatography. Methods Outer environment water incubated in alkaline peptone water(36.5℃)for 6 hours. Then vibrio cholerae was detected by colloidal gold method. The traditional separation culture method of vibrio cholerae was identified with serum agglutina as the gold standard at the same time. The results of two methods were compared by statistical methods. Results Among 747 water samples, 19 were identified with colloidal gold and 16 vibrio cholerae were identified by the traditional separation culture method. There was no significant difference between the two methods(χ^2=0.61, P〈0.05). For the colloidal gold method to detect vibrio cholerae of outer environment water, the sensitivity was 100.00%, with a specificity of 99.59%,a false positive rate of 0.41%, a false negative rate of 0.00% and a consistent rate of 99.60% with the traditional separation culture method. Conclusion The colloidal gold method has good sensitivity and specificity and consistency with traditional separation culture method for detection of vibrio cholerae in outer environment water. The method of detecting vibrio cholerae with colloidal gold is reliable, simple, practical and suitable for use at grassroots level as an auxiliary method.
