摘 要:目的:探讨银杏酮酯(ginkgo biloba extract 50,GBE50)联合芹菜素(apigenin)预处理对大鼠心肌缺血再灌注损伤的保护作用。方法:建立大鼠心肌缺血再灌注损伤模型,选用Sprague-Dawley(SD)大鼠50只随机分为正常组、模型组、银杏酮酯组、芹菜素组、银杏酮酯联合芹菜素组,均按照100 mg/kg灌胃。检测血浆丙二醛(MDA)、肌酸激酶同工酶(CK-MB)、肌钙蛋白T(c Tn T)、肌酸激酶(CK)和心脏组织超氧化物歧化酶(SOD)和白介素-6(IL-6)变化。同时,取心脏组织HE染色,观察心肌组织的病理变化。结果:与模型组相比,银杏酮酯组100mg/kg、芹菜素组100mg/kg能降低MDA、CK-MB、c Tn T、CK、IL-6水平和升高SOD水平;与银杏酮酯组100mg/kg、芹菜素组100mg/kg相比,银杏酮酯联合芹菜素组100mg/kg能显著降低MDA、CK-MB、c Tn T、CK、IL-6水平和升高SOD水平;与模型组相比,银杏酮酯组100mg/kg、芹菜素组100mg/kg能减轻心肌组织损伤的病理学变化;与银杏酮酯组100mg/kg、芹菜素组100mg/kg相比,银杏酮酯联合芹菜素组100mg/kg能显著减轻心肌组织损伤的病理学变化。结论:与银杏酮酯100mg/kg、芹菜素100mg/kg相比,银杏酮酯联合芹菜素100mg/kg对大鼠心肌缺血再灌注损伤具有明显的保护作用,这可能与提高心脏组织中SOD水平和降低心脏组织中IL-6水平有关。Objective: To study the protective effect of Ginkgo biloba extract 50 joint Apigenin on heart ischemia / reperfusion( I / R) injury in rats.Methods: The model was made by ligating the left desceding coronary artery for 45 minutes and reperfusing for 2 hour in rat. 50 rats were divided into control group,ischemia group,Ginkgo biloba extract 50 group 100 mg / kg,Apigenin group 100 mg / kg,Ginkgo biloba extract 50 joint Apigenin group 100 mg / kg. Ginkgo biloba extract 50 joint Apigenin were used on the rats at the beginning of two weeks. Plasma MDA,CK- MB,c Tn T,CK and tissue SOD,IL-6 were observed. Myocardial injury degree was observed though hitopathological biopsy. Results:MDA,CK- MB,c Tn T,CK,IL-6 were lower and SOD was higher of Ginkgo biloba extract 50 group 100 mg / kg and Apigenin group 100 mg / kg than those ischemia group( P 〈 0. 05). MDA,CK- MB,c Tn T,CK,IL-6 were significantly lower and SOD was significantly higher of Ginkgo biloba extract 50 joint and Apigenin group 100 mg / kg than Ginkgo biloba extract 50 group 100 mg / kg and Apigenin group 100 mg / kg( P 〈 0.05). Pathological damage of myocardial cells in Ginkgo biloba extract 50 group 100 mg / kg and Apigenin group 100 mg / kg were lighter than that of ischemia group. Pathological damage of myocardial cells in Ginkgo biloba extract 50 joint Apigenin group 100 mg / kg was significantly lighter than those of Ginkgo biloba extract 50 group 100 mg / kg and Apigenin group 100 mg / kg. Conclusion: Our results indicate that Ginkgo biloba extract 50 joint Apigenin 100 mg / kg have a significantly protective effect on heart ischemia / reperfusion injury in rats than Ginkgo biloba extract 50 group 100 mg / kg and Apigenin group 100 mg / kg because it can increase SOD in heart tissue,decrease IL-6 in heart tissue.
