安息香对孕妊大鼠血清性激素、在体子宫平滑肌的影响

摘　　要：目的:观察安息香对雌性孕妊大鼠血清雌二醇、孕酮以及在体子宫平滑肌的影响。方法:依据阴栓观察法和阴道涂片法确定大鼠孕期;通过酶联免疫吸附分析法(ELISA)测定血清雌二醇(E2)及孕酮(PROG)水平,称重法测定子宫和卵巢指数;采用生物机能系统测定不同浓度安息香对孕妊大鼠在体子宫平滑肌张力、频率和峰面积的影响;并观测安息香1.0g/kg对缩宫素、米索前列醇对子宫平滑肌的干预机制。结果:安息香0.5、1.0g/kg组均能显著增加孕妊大鼠血清中雌二醇、孕酮水平,增加卵巢和子宫指数,并能显著抑制在体子宫平滑肌的张力、频率和峰面积;安息香1.0 g/kg能显著对抗缩宫素和米索前列醇后子宫平滑肌的收缩张力、平均面积、频率;再次加入缩宫素能明显增强安息香后的子宫平滑肌的张力和收缩频率;再次加入米索前列醇,对安息香作用后的子宫平滑肌则无影响。结论:安息香能增加孕妊大鼠血清中雌性激素水平,同时可能通过阻断孕妊大鼠在体子宫平滑肌的前列腺受体和部分阻断缩宫素受体而抑制子宫平滑肌运动。Objective： This paper aims to study the effect of benzoinum on sex- hormone and to observe the effect and its mechanisms of benzoinum on uterine smooth muscle in health female pregnant rats. Methods： Observeing the estrous cycle changes of each group by vaginal exfoliated cells to ensure the pregnancy; estradiol（ E2） and progesterone（ P） in serum was measured with enzyme linked immunosorbent assay（ELISA）; the index of uterine and ovarian was calculated by weigting rat’s uterus and ovary; and the muscle tension,frequency and peak areas of early pregnant rats uterine smooth muscle in vivo of benzoinum in different dosages by using biological functional system and observation the intervention mechanisms of the uterine smooth muscle of benzoin of 1. 0g / kg on oxytocin and misoprostol. Results： The doses of benzoinum0. 5,1. 0g / kg could not only significantly higher the level of E2 and P but also significanty increase the index of uterus and ovary（ P 〈 0. 05）,meanwhile,the benzoinum could remarkably decrease the tension and peak area as well as frequency of pregnant rats uterine smooth muscle（ P 〈 0. 05）; The dosages of benzoinum 1g/kg,significantly against the uterus after misoprostol and oxytocin of the tension and peak area as well as frequency of pregnant rats uterine smooth muscle（ P 〈 0. 05）; rejoined oxytocin could significantly increase the tension and frequency of pregnant rats uterine smooth muscle after benzoin（ P 〈 0. 05）; And once again joined the misoprostol no significant effects on uterine smooth muscle after benzoin（ P 〉 0. 05）. Conclusion： Continuous use of benzoinum with high dosage（ 1g / kg） not only could significantly higher the level of E2 and P in serum but also increase the development of reproductive organs,and it also likely works by blocking receptors of the prostate and partly OT receptors to inhibit the activity of the uterine smooth mucle in pregnant rats.
