小儿解表颗粒解热镇痛抗炎作用研究

摘　　要：目的：考察小儿解表颗粒的解热、镇痛及抗炎效果及其抗炎机制,为其临床应用提供依据。方法：通过内毒素致家兔发热的模型观察解热作用;小鼠热板法观察镇痛作用;角叉菜胶诱导大鼠足肿胀观察抗炎作用;ELISA法检测小儿解表颗粒对大鼠血清的细胞因子含量IL-1β、IL-6、IL-10的影响。结果：1解热试验：与模型组相比,小儿解表3.36g/kg、1.68g/kg组分别在2 h、5.5h时对发热家兔体温表现出抑制作用,且3.36g/kg、1.68g/kg组在5.5h降温效果优于布洛芬组。2镇痛实验：与模型组相比,小儿解表9.6g/kg、2.4g/kg组分别在1.5h、3h痛阈提高百分率有显著增加。3抗炎实验：与模型组相比,15.36g/kg、7.68g/kg组分别在3~4h对大鼠足肿胀作用呈明显抑制作用;而与布洛芬组相比,15.36g/kg、7.68g/kg组均无显著差异。4ELISA实验：与模型组相比,小儿解表15.36g/kg、7.68g/kg组大鼠血清细胞因子IL-1β表达量明显降低,15.36g/kg组大鼠血清细胞因子IL-6表达量明显降低;15.36g/kg组大鼠血清细胞因子IL-10表达量明显增加。结论：小儿解表颗粒具有一定的解热、镇痛及抗炎作用,其机制可能与下调血清细胞因子IL-1β、IL-6的水平和上调细胞因子IL-10的水平等相关细胞因子的表达有关。Objective： To study the antipyretic,analgesic,anti-inflammatory reaction of Xiaoerjiebiao granules and explore its mechanisms,and provides the theory basis for its clinical application. Methods： The febrile model of rabbits induced by bacterial endotoxin,hot-plate test in mice,The rats model of toes swelling caused by carrageenan were used to observe the antipyretic,analgesic,anti-inflammatory actions of Xiaoerjiebiao granules; and the potential mechanism of xiaoerjiebiao granules were studied through detecting the concentration of Interleukin-1β（ IL-1β）,Interleukin-6（ IL-6）,Interleukin-10（ IL-10） in serum by ELISA. Results： 1 Antipyretic experiment： Compared with the control group,the heating temperature of rabbits in 1. 68 g / kg,3. 36 g / kg group at 2h,5. 5h showed inhibitory effect（ p 〈 0. 05）; the cooling effect in1. 68 g / kg,3. 36 g / kg group at 5. 5h was better than that of the ibuprofen group. 2 Analgesic experiment： compared with the control group,the pain threshold improved of mice in 9. 6g / kg,2. 4g / kg group respectively at 1. 5h,3h was increased significantly（ p 〈 0. 05）. 3Anti-inflammatory experiment： compared with the control group,15. 36 g / kg,7. 68 g / kg group of Xiaoerjiebiao granules respectively in the 3 ~ 4h could remarkably inhibit paw edema in rats（ P 〈 0. 05）; compared to ibuprofen group,there were no significant differences in 15. 36 g / kg,7.68 g / kg group. 4 ELISA experiment： compared with the control group,rats serum cytokine expression of IL-1 of Xiaoerjiebiao granules in15. 36 g / kg,7. 68 g / kg group were significantly decreased（ P 〈 0. 05）; rats serum cytokines expression of IL-6 in 15. 36 g / kg group were significantly decreased（ P 〈 0. 05）; rats serum cytokine expression of IL-10 in 15. 36 g / kg group were significantly increased（ P 〈 0. 05）.
