经侧脑室注射芪苈强心胶囊对慢性心衰大鼠的影响

摘　　要：目的:观察经微量注射泵给予慢性心衰大鼠侧脑室芪苈强心胶囊对其心功能、心肌组织形态、神经内分泌因子及下丘脑的影响。方法:采用结扎大鼠冠状动脉左前降支建立心衰模型,造模4周后通过侧脑室插管连接微量注射泵给予芪苈强心胶囊溶液(分为0.01mg/kg、0.02mg/kg剂量组),阳性对照组给予CRH竞争性抑制剂(αh-CRH),给药4周后,应用颈动脉插管检测各组大鼠心功能指标,HE染色观察心肌组织形态,酶联免疫吸附试验检测大鼠血清N-端脑钠肽前体(NT-pro BNP)与血浆促肾上腺皮质激素(ACTH)、前列腺素E2(PGE2)、去甲肾上腺素(NE)含量,RT-PCR方法检测下丘脑促肾上腺皮质激素释放激素(CRH)mRNA与肿瘤坏死因子-α(TNF-α)mRNA的表达。结果:芪苈强心胶囊溶液0.01mg/kg组、芪苈强心胶囊溶液0.02mg/kg组与CRH竞争性抑制剂组均可改善慢性心衰大鼠心功能,减轻心肌组织形态的改变,降低大鼠血清NT-pro BNP与血浆ACTH、PEG2、NE含量,降低下丘脑CRH mRNA与TNF-αmRNA的表达。结论:经侧脑室插管连接微量注射泵给予芪苈强心胶囊可有效改善心衰大鼠心功能和心肌组织形态的改变,降低神经内分泌因子的含量,并且降低下丘脑CRH mRNA与TNF-αmRNA的表达,对心衰大鼠产生干预影响。Objective： To explore the influence of traditional Chinese medicine Qiliqiangxin given throw the lateral ventricle on chronic heart failure rats cardiac function,myocardial tissue morphology,neuroendocrine factors and hypothalamic. Methods： The heart failure rat model was established by the left anterior descending coronary artery ligation. The rats were given Qiliqiangxin solution（ 0. 01 mg / kg and 0. 02 mg / kg）,and positive drug- CRH competitive inhibitor（ αh- CRH） by the micro injection pump to the lateral ventricle for four weeks after model.After four weeks,blood flow dynamics indicators were measured by carotid artery intubation,and HE was used to observe myocardial tissue morphology,and Enzyme linked immunosorbent assay was used to measure serum NT- pro BNP and plasma adrenocorticotropic hormone（ ACTH）,prostaglandin E2（ PEG2） and norepinephrine（ NE）,and PCR was used to detect the hypothalamus CRH mRNA and TNF-αmRNA. Results： Qiliqiangxin solution group and positive drug group can improve cardiac function,lower serum NT- pro BNP and plasma ACTH,PEG2 and NE,and can reduce the expression of the hypothalamus CRH mRNA and TNF- αmRNA in rats with heart failure. Conclusion： Giving traditional Chinese medicine to the lateral ventricles can improve cardiac function,reduce the myocardial tissue morphology changes and the blood levels of neuroendocrine factors,lower the expression of the hypothalamus CRH mRNA and the pituitary TNF-αmRNA,and have a certain effect on the changes in heart failure rats.
